Janelia Farm grup lideri Eric Betzig \u201cE\u011fer biri oyunun kurallar\u0131n\u0131 \u00f6\u011frenmeye \u00e7abal\u0131yorsa, bu oyunun foto\u011fraflar\u0131na bakmak yerine o oyunu oynayan insanlar\u0131n bir filmini izlemesi daha avantajl\u0131d\u0131r. Betzig ayn\u0131 zamanda \u201cDura\u011fan resimler gibi olan \u00f6l\u00fc h\u00fccrelerden \u00f6\u011frenilebilecek \u015feyler s\u0131n\u0131rl\u0131d\u0131r\u201d diyor.<\/p>\n<p>Betzig, ara\u015ft\u0131rmac\u0131lara canl\u0131 h\u00fccrelerin i\u00e7 ya\u015fam\u0131n\u0131 g\u00f6rme olana\u011f\u0131 sa\u011flayan bir mikroskop yaratmak istemi\u015f. Floresan molek\u00fcllerle i\u015faretlenmi\u015f protein ve di\u011fer molek\u00fcllerin olu\u015fturdu\u011fu noktalarca resmedilmi\u015f canl\u0131 h\u00fccrelerle \u00e7al\u0131\u015fma fikri yeni de\u011fil. Fakat canl\u0131-h\u00fccre teknikleri, mikroskop taraf\u0131ndan \u00fcretilen \u0131\u015f\u0131k zamanla h\u00fccreye zarar verebilece\u011fi i\u00e7in problemli olabilir. O, (k\u0131r\u0131n\u0131m yasalar\u0131 taraf\u0131ndan dayat\u0131lan) uzaysal \u00e7\u00f6z\u00fcnmenin limitlerine zarar verecek olan yeni mikroskopi tekniklerini tasarlamaya odaklanm\u0131\u015fken bu d\u00fc\u015f\u00fcncelerini bir s\u00fcre saklam\u0131\u015f. Bu yol \u0131\u015f\u0131\u011f\u0131n numuneye zarar vermesine ve bulan\u0131kl\u0131\u011fa neden olur, bu y\u00fczden canl\u0131 h\u00fccreleri g\u00f6zlemlemeyi zorla\u015ft\u0131r\u0131r.<\/p>\n<p>2008 y\u0131l\u0131nda Betzig bu zorlu\u011fun \u00fcstesinden gelmenin yollar\u0131 \u00fczerinde \u00e7al\u0131\u015fmaya ba\u015flam\u0131\u015f. Sahip oldu\u011fu fikirlerden biri de d\u00fczlem ayd\u0131nlatma mikroskopisini kullanmak. \u0130lk olarak 100 y\u0131l kadar \u00f6nce \u00f6nerilen d\u00fczlem ayd\u0131nlatmas\u0131, numunenin \u00fcst k\u0131sm\u0131 yerine yan y\u00fcz\u00fcnden gelen parlayan bir \u0131\u015f\u0131k katman\u0131 i\u00e7erir. Bunu yapmak i\u00e7in mikroskop uzmanlar\u0131 birbirlerine dik iki farkl\u0131 objektif lens kullan\u0131rlar. Betzig durumu: \u201cYan k\u0131s\u0131mlardan geldi\u011finiz i\u00e7in d\u00fczlem ayd\u0131nlatma, odaklanan k\u0131sma \u00e7ok yak\u0131n olan eksitasyonu (uyar\u0131lma) s\u0131n\u0131rlar\u201d \u015feklinde a\u00e7\u0131kl\u0131yor.<\/p>\n<p>D\u00fczlem ayd\u0131nlatma, Janelia Farm akademi \u00fcyesi Philipp Keller de dahil olmak \u00fczere di\u011fer ara\u015ft\u0131rmac\u0131lar taraf\u0131ndan, y\u00fczlerce mikron boyutlar\u0131ndaki \u00e7ok h\u00fccreli organizmalar\u0131 incelerken etkiyi art\u0131rmada kullan\u0131lmas\u0131na ra\u011fmen, \u0131\u015f\u0131k katmanlar\u0131 sadece 10 mikron boyutundaki h\u00fccrelerin i\u00e7ini efektif \u015fekilde g\u00f6r\u00fcnt\u00fclemek i\u00e7in h\u00e2l\u00e2 \u00e7ok kal\u0131nd\u0131. As\u0131l problem, Betzig\u2019in grubunun istedi\u011finden daha fazla h\u00fccrenin d\u00fczlem ayd\u0131nlatmadaki geni\u015f \u0131\u015f\u0131k \u015feritlerine maruz kalmas\u0131yd\u0131. Bu durum a\u015f\u0131r\u0131 bulan\u0131kl\u0131\u011fa ve hafif toksisiteye sebep oluyordu. Bu problemi a\u015fmak i\u00e7in Betzig ve grubu, k\u0131r\u0131n\u0131ma u\u011framayan \u00f6zel tip \u0131\u015f\u0131k demeti olan Bessel \u0131\u015f\u0131nlar\u0131n\u0131 kullanm\u0131\u015f. Bessel \u0131\u015f\u0131nlar\u0131, 1980\u2019lerin sonunda fizik\u00e7iler taraf\u0131ndan incelenmi\u015f ve bug\u00fcn s\u00fcpermarketlerde kullan\u0131lan barkod tarama cihazlar\u0131n\u0131 da i\u00e7eren uygulamalarda kullan\u0131l\u0131yor. B\u00f6ylece iki fotonlu metot uyguland\u0131\u011f\u0131nda Bessel taraf\u0131ndaki loplar\u0131n arka plan\u0131 elenir ve geriye kalanlar da Bessel \u0131\u015f\u0131nlar\u0131n\u0131n dar merkez b\u00f6l\u00fcm\u00fcndeki \u0131\u015f\u0131kt\u0131r.<\/p>\n<p>Daha sonra olabildi\u011fince h\u0131zl\u0131 bir \u015fekilde g\u00f6r\u00fcnt\u00fcleme yapmaya koyulmu\u015flar.
