Art\u0131k yakla\u015f\u0131k 2 metre uzunlu\u011fundaki genomun boyu 1 milimetrenin y\u00fczde biri kadar olan tek bir h\u00fccre \u00e7ekirde\u011fine s\u0131\u011fabilmek i\u00e7in ne \u015fekilde katland\u0131\u011f\u0131na y\u00f6nelik bir fikrimiz var.<\/p>\n<p>MIT (Massachusetts Institute of Technology),\u00a0 Harvard ve MIT ortakl\u0131\u011f\u0131 i\u00e7erisinde bulunan Broad Institute ve Harvard T\u0131p Okulu\u2019nda yap\u0131lan ortak bir \u00e7al\u0131\u015fmayla insan genomunun 3 boyutlu yap\u0131s\u0131 ortaya \u00e7\u0131kar\u0131ld\u0131. B\u00f6ylelikle 3 milyar baz \u00e7iftinin bir h\u00fccre \u00e7ekirde\u011fine s\u0131\u011fabilmek i\u00e7in nas\u0131l organize oldu\u011fu bilinenler aras\u0131na eklendi.<\/p>\n<p><em>Science<\/em> dergisinde yay\u0131mlanan \u00e7al\u0131\u015fma ayn\u0131 zamanda DNA\u2019n\u0131n baz\u0131 k\u0131s\u0131mlar\u0131 aktif \u015fekilde okunurken baz\u0131 k\u0131s\u0131mlar\u0131n\u0131n da nas\u0131l \u201csessiz\u201d kald\u0131\u011f\u0131n\u0131 a\u00e7\u0131klayabilir. Dahas\u0131, geli\u015ftirilen bu yeni teknik yard\u0131m\u0131yla ara\u015ft\u0131rmac\u0131lar, gen ekspresyonunun h\u00fccreler geli\u015firken veya kanserle\u015firken nas\u0131l de\u011fi\u015fti\u011fi anla\u015f\u0131labilecek. Kromozomlar\u0131n h\u00fccre i\u00e7indeki yerlerini ve nas\u0131l pozisyonland\u0131klar\u0131n\u0131 g\u00f6sterebilen bu teknik sayesinde DNA\u2019daki bilginin fonksiyonel anlamda nas\u0131l d\u00fczenlendi\u011fini de anlama olana\u011f\u0131 bulaca\u011f\u0131z.<\/p>\n<p>Yap\u0131sal veriler insan genomunun, aktif genler eri\u015filebilir bulunurken, kullan\u0131lmayan DNA b\u00f6lgesinin daha yo\u011fun b\u00f6lgelere s\u0131k\u0131\u015ft\u0131\u011f\u0131 iki ayr\u0131 kompart\u0131mana ayr\u0131larak organize oldu\u011funu g\u00f6steriyor.<\/p>\n<p>\u00c7al\u0131\u015fman\u0131n sonu\u00e7lar\u0131na g\u00f6re her kromozom, aktif gen-zengini DNA b\u00f6lgeleri ve inaktif gen-fakiri b\u00f6lgeleri bu mant\u0131kla organize ediyor.<\/p>\n<p>\u00c7al\u0131\u015fma i\u00e7in MIT\u2019deki fizik\u00e7iler ile yap\u0131lan i\u015fbirli\u011fi sonucunda DNA\u2019n\u0131n matematikte bilinen \u201cfraktal\u201d \u015feklinde oldu\u011fu g\u00f6r\u00fcld\u00fc. Bu mimari, h\u00fccrenin DNA\u2019daki d\u00fc\u011f\u00fcmler ve katlanmalar\u0131 \u00f6nleyerek genomu okuyabilmesini olanakl\u0131 k\u0131lacak \u201cfraktal glob\u00fcl\u201d olarak adland\u0131r\u0131l\u0131yor. Ayr\u0131ca DNA\u2019n\u0131n gen aktivasyonu, bask\u0131lanmas\u0131 ve h\u00fccre b\u00f6l\u00fcnmesi s\u00fcre\u00e7lerinde kolayl\u0131kla a\u00e7\u0131l\u0131p kapanabilmesini sa\u011fl\u0131yor.<\/p>\n<p>Do\u011fan\u0131n bilgiyi saklamas\u0131n\u0131n zarif bir bi\u00e7imi s\u00fcper-yo\u011fun, d\u00fc\u011f\u00fcms\u00fcz bir fraktal. Fakat bu yap\u0131n\u0131n kolayl\u0131kla d\u00fc\u011f\u00fcmlenebilir oldu\u011fu ve fiziksel olarak zor hareket edebilmesiyle ilgili saptamalarda bulundular.<\/p>\n<p>Genomun yap\u0131s\u0131n\u0131 de\u015fifre etmeye yarayan Hi-C tekni\u011fi, tekil olarak genlerin birbirine yak\u0131nl\u0131\u011f\u0131n\u0131 inceleyebiliyor. \u00c7al\u0131\u015fmay\u0131 ger\u00e7ekle\u015ftirenler \u00f6nce h\u00fccre \u00e7ekirde\u011finde bulunan DNA ipliklerini bir arada tutabilmek i\u00e7in formaldehit kulland\u0131lar. Daha sonra DNA\u2019y\u0131 par\u00e7alay\u0131p k\u00fc\u00e7\u00fck par\u00e7alara ay\u0131rarak kom\u015fu segmentleri belirlediler. DNA k\u00fc\u00e7\u00fck halkac\u0131klar halinde tekrar birle\u015ftirildikten sonra paralel DNA sekanslama a\u015famas\u0131na ge\u00e7ildi.<\/p>\n<p>Liberman ve di\u011fer e\u015f yazar Aiden elde ettikleri verinin bir fraktal glob\u00fcl olu\u015fturdu\u011funu g\u00f6sterdiler.
