Bu g\u00fc\u00e7lenmenin uzun s\u00fcreli potansiyel art\u0131\u015f\u0131 (long-term potentiation (LTP)) olarak isimlendirilen sinirsel esnekli\u011fin (sinaptik plastisite) bir bi\u00e7imiyle ger\u00e7ekle\u015fti\u011fi varsay\u0131l\u0131yordu, ne yaz\u0131k ki bu konuda sa\u011flam bir kan\u0131t mevcut de\u011fildi.<\/p>\n<p>Yeni ara\u015ft\u0131rma geli\u015fen n\u00f6roteknolojiyi kullanarak sadece korku haf\u0131zas\u0131n\u0131n olu\u015fumu i\u00e7in LTP ile sinapslar\u0131n g\u00fc\u00e7lendirilmesinin gerekti\u011fini g\u00f6stermekle kalm\u0131yor, ayn\u0131 zamanda bu haf\u0131zan\u0131n ayn\u0131 sinapslar\u0131 zay\u0131flatarak silinebildi\u011fini de g\u00f6steriyor.<\/p>\n<p>LTP ilk defa 60\u2019larda Norve\u00e7li bilim insan\u0131 Terje L\u00f8mo taraf\u0131ndan ke\u015ffedildi. L\u00f8mo tav\u015fan hipokampusundaki sinir h\u00fccrelerinin etkinli\u011fini s\u0131namak i\u00e7in elektrot \u00e7iftleri kullan\u0131yordu. L\u00f8mo bir elektrotu bir b\u00f6lgedeki h\u00fccreleri uyarmak i\u00e7in ve bir di\u011ferini ise kom\u015fu alandaki ilintili h\u00fccrelerin etkinli\u011fini e\u015f zamanl\u0131 \u00f6l\u00e7mek i\u00e7in kulland\u0131. L\u00f8mo elektrikle uyar\u0131lan h\u00fccrelerin aralar\u0131ndaki ba\u011flant\u0131n\u0131n g\u00fc\u00e7lendi\u011fini g\u00f6zledi. \u00d6zellikle bir h\u00fccre di\u011feriyle \u00f6rt\u00fc\u015fecek bi\u00e7imde d\u00fczenli ve tekrarlayan y\u00fcksek frekansl\u0131 uyar\u0131ya maruz b\u0131rak\u0131ld\u0131\u011f\u0131nda aralar\u0131ndaki iletim daha verimli hale geliyordu, k\u00fc\u00e7\u00fck bir ate\u015fleme akabinde di\u011ferinde uzat\u0131lm\u0131\u015f bir tepki \u00fcretiyordu.<\/p>\n<p>Hipokamp\u00fcs\u00fcn haf\u0131za i\u00e7in \u00f6nemi o zamana kadar iyice anla\u015f\u0131lm\u0131\u015ft\u0131, ard\u0131ndan LTP ke\u015ffedildi, LTP\u2019nin haf\u0131zan\u0131n h\u00fccresel altyap\u0131s\u0131 oldu\u011fu yayg\u0131n \u015fekilde \u00f6ng\u00f6r\u00fcld\u00fc ve beyin ara\u015ft\u0131rmalar\u0131ndaki di\u011fer alanlara k\u0131yasla daha fazla makale \u00fcretilerek mekanizmas\u0131 hakk\u0131nda yo\u011fun bi\u00e7imde \u00e7al\u0131\u015f\u0131ld\u0131. Bu \u00e7al\u0131\u015fmalar\u0131n pek \u00e7o\u011fu LTP\u2019nin \u00e7e\u015fitli haf\u0131za bi\u00e7imleriyle ili\u015fkili oldu\u011funu, \u00e7e\u015fitli kimyasallarla veya genetik mutasyonlarla mekanizman\u0131n bloke edilmesinin haf\u0131za kay\u0131plar\u0131na yol a\u00e7t\u0131\u011f\u0131n\u0131 ortaya koydu. Ancak hi\u00e7biri haf\u0131zan\u0131n h\u00fccresel d\u00fczeyde LTP\u2019ye dayand\u0131\u011f\u0131n\u0131 do\u011frudan g\u00f6stermiyordu. Roberto Malinow ve meslekta\u015flar\u0131 bu probleme optogenetik olarak isimlendirilen bir teknikle yakla\u015ft\u0131lar. Bu teknik Channelrhodopsin olarak adland\u0131r\u0131lan \u0131\u015f\u0131\u011fa duyarl\u0131 proteini kodlayan genin se\u00e7ilen sinir h\u00fccresi gruplar\u0131na aktarmay\u0131 i\u00e7eriyor.
