Ancak biliminsanlar\u0131, h\u00fccreleri yeniden programlayacak olan bu i\u015flemin, h\u00fccrelerin kullan\u0131\u015fl\u0131l\u0131\u011f\u0131n\u0131 k\u0131s\u0131tlayan baz\u0131 genetik anormallikleri ba\u015flatabilece\u011fini fark etmi\u015flerdi.<\/p>\n<p>IPSCs \u00fcretebilmek i\u00e7in yeti\u015fkin h\u00fccreler, embriyonik k\u00f6k h\u00fccrelerde aktif halde bulunan genlerle olu\u015fturulmu\u015f bir kokteyle maruz b\u0131rak\u0131ld\u0131. Daha sonra bu k\u00f6k h\u00fccreler b\u00f6l\u00fcnerek sinir ve kas gibi di\u011fer h\u00fccre tiplerine ayr\u0131ld\u0131. Hebrew \u00dcniversitesi T\u0131p Fak\u00fcltesi\u2019nde bulunan \u0130srail-Kanada T\u0131bbi Ara\u015ft\u0131rma Enstit\u00fcs\u00fc\u2019nden Dr. Yosef Burganim, MIT\u2019de biyoloji profes\u00f6r\u00fc olan Rudolf Jaenisch\u2019in kurucu \u00fcyesi oldu\u011fu Whitehead Enstit\u00fcs\u00fc Laboratuvar\u0131n\u0131n biliminsanlar\u0131yla \u00e7al\u0131\u015f\u0131yor.<\/p>\n<p>Ara\u015ft\u0131rmac\u0131lar, yeniden programlay\u0131c\u0131 etkenlerin de\u011fi\u015ftirilmesinin, h\u00fccrelerin yenilenmesinde daha kontroll\u00fc ve y\u00fcksek kaliteli IPSCs kazand\u0131racak bir yol oldu\u011fu fikrine vard\u0131lar. Bunun i\u00e7in fare h\u00fccreleri \u00fczerinde \u00e7al\u0131\u015fan Dr. Burganim ve ara\u015ft\u0131rmac\u0131 biliminsan\u0131 Styliani Markoulaki, yeni etken kokteylini tasarlamak i\u00e7in biyobili\u015fim analizi kulland\u0131lar (Yeni etkenler Sall4, Nanog, Esrrb ve Lin28, k\u0131saca SNEL\u2019dir).<\/p>\n<p>Sonu\u00e7lar, yeniden programlay\u0131c\u0131 etkenler aras\u0131ndaki etkile\u015fimin elde edilen IPSCs say\u0131s\u0131 ve kalitesinde olduk\u00e7a \u00f6nemli bir rol oynad\u0131\u011f\u0131n\u0131 g\u00f6sterdi; bamba\u015fka bir etken birle\u015fimiyle daha kaliteli \u00fcr\u00fcnler elde edilecek olabilir.<\/p>\n<p>Yeni SNEL kokteyli daha az IPSCs kolonisi \u00fcretiyor, fakat h\u00fccrelerin yakla\u015f\u0131k olarak y\u00fczde 80\u2019i en zor pluripotent testlerini ba\u015far\u0131yla ge\u00e7ebiliyor. Bu yeni kokteyl, daha fazla say\u0131da k\u00f6k h\u00fccre kolonisi \u00fcreten, ancak b\u00fcy\u00fck \u00e7o\u011funlu\u011funun pluripotent testlerini ge\u00e7emedi\u011fi geleneksel OSKM kokteyline g\u00f6re \u00e7ok daha tercih edilebilirdir.
