Biliminsanlar\u0131 bu molek\u00fcllerin D\u00fcnyada ve di\u011fer gezegenlerde basit karbon temelli canl\u0131l\u0131\u011f\u0131n ortaya \u00e7\u0131kmas\u0131ndan sorumlu oldu\u011funu d\u00fc\u015f\u00fcn\u00fcyor. Fakat insanlar gibi, daha geli\u015fmi\u015f ve DNA temelli kompleks canl\u0131lar i\u00e7in BaP b\u00fcy\u00fck bir \u00e7evresel tehdit olu\u015fturabiliyor. Fakat hi\u00e7biri sigara i\u00e7mek kadar insan v\u00fccuduna BaP girmesine sebep olmuyor.<\/p>\n<p>Genellikle e\u011fer toksik bir hidrokarbon insan v\u00fccuduna solunum veya beslenme yoluyla girerse, kan\u0131m\u0131zdaki baz\u0131 enzimler bu molek\u00fclleri daha k\u00fc\u00e7\u00fck ve daha tehlikesiz molek\u00fcllere par\u00e7alarlar. Bu BaP i\u00e7in de ge\u00e7erli, fakat bu sefer koruyucu ama\u00e7l\u0131 mekanizma benzo[\u03b1]piren diol epoksid (BPDE) olarak adland\u0131r\u0131lan ve asl\u0131nda BaP\u2019\u0131n kendisinden daha zararl\u0131 olan bir yan \u00fcr\u00fcn olu\u015fturuyor.<\/p>\n<p>BPDE, DNA ile kimyasal reaksiyona giriyor ve DNA\u2019daki guanin bazlar\u0131 ile \u00e7ok s\u0131k\u0131 bir ba\u011f kuruyor. Bu ba\u011flar, ba\u015fka bir deyi\u015fle\u00a0 DNA-kat\u0131m\u0131 ise (DNA adducts), DNA\u2019n\u0131n d\u00fczg\u00fcn proteinler \u00fcretebilmesini ve h\u00fccre b\u00f6l\u00fcnmesi s\u0131ras\u0131nda d\u00fczg\u00fcnce e\u015flenebilmesini engelliyor. Bunun sonucunda da hastal\u0131klar ortaya \u00e7\u0131k\u0131yor.<\/p>\n<p>Bu \u00e7al\u0131\u015fman\u0131n birinci yazar\u0131 olan Dr. Wentao Li \u015f\u00f6yle s\u00f6yl\u00fcyor: \u201cE\u011fer BPDE bir t\u00fcm\u00f6r bast\u0131r\u0131c\u0131 (<em>tumor suppresor<\/em>) gen \u00fczerinde bozulmaya sebep olursa, bu durum kal\u0131c\u0131 mutasyonlara yol a\u00e7ar ve h\u00fccre bir kanser h\u00fccresine d\u00f6n\u00fc\u015f\u00fcr.\u201d<\/p>\n<p>Bu kimyasal reaksiyonun kansere sebep oldu\u011fu konusunda bir \u015f\u00fcphe yok. BaP, BPDE dolay\u0131s\u0131yla \u00e7ok uzun s\u00fcredir pek \u00e7ok kanser t\u00fcr\u00fcn\u00fcn tetikleyici ve akci\u011fer kanserinin tek ve en g\u00fc\u00e7l\u00fc sebebi olarak kabul edilmektedir.<\/p>\n<p><strong>Onar\u0131m a\u015famas\u0131<\/strong><\/p>\n<p>Sancar\u2019\u0131n BaP kaynakl\u0131 DNA hasar\u0131n\u0131n haritalanmas\u0131n\u0131 sa\u011flayan y\u00f6ntemi, biliminsanlar\u0131n\u0131n h\u00fccre i\u00e7erisinde DNA hasar\u0131n\u0131n onar\u0131lmaya \u00e7al\u0131\u015f\u0131ld\u0131\u011f\u0131 noktalar\u0131 saptamas\u0131na olanak sa\u011flayacak. Sancar daha \u00f6nce bu DNA onar\u0131m\u0131n\u0131n biyokimyasal basamaklar\u0131n\u0131n detayl\u0131ca ayd\u0131nlat\u0131lmas\u0131nda rol alarak 2015 Nobel Kimya \u00f6d\u00fcl\u00fcn\u00fc payla\u015ft\u0131.<\/p>\n<p>N\u00fckleotid eksizyon onar\u0131m\u0131 olarak adland\u0131r\u0131lan bu onar\u0131m mekanizmas\u0131, DNA\u2019n\u0131n kesilip bi\u00e7ilmesinden, bir nevi\u00a0 DNA cerrahl\u0131\u011f\u0131ndan sorumlu \u00f6zel proteinlerin takviyesini i\u00e7eriyor. Bu proteinler DNA\u2019n\u0131n hasarl\u0131 par\u00e7alar\u0131n\u0131 k\u0131rp\u0131yorlar. E\u011fer bu s\u00fcre\u00e7 ba\u015far\u0131l\u0131 tamamlan\u0131rsa, DNA sentezleyen enzimler, bu bo\u015flu\u011fu hasarl\u0131 olmayan DNA e\u015fini baz alarak dolduruyor.\u00a0 Bu, b\u00fct\u00fcn h\u00fccre temelli ya\u015fam formlar\u0131n\u0131n 2 komplemanter DNA ipli\u011fi bulundurmas\u0131 sayesinde m\u00fcmk\u00fcn olabiliyor. Sonu\u00e7 olarak biliminsanlar\u0131 DNA onar\u0131m\u0131n\u0131n ba\u015flad\u0131\u011f\u0131 par\u00e7alar\u0131n tamamlanm\u0131\u015f bir haritas\u0131n\u0131 elde etmeyi ba\u015fard\u0131.<\/p>\n<p>Y\u00fcksek maliyeti ve \u00e7ok emek gerektirmesi sebebiyle Sancar, Li ve meslekta\u015flar\u0131n\u0131n yay\u0131mlad\u0131klar\u0131 ilk harita m\u00fcmk\u00fcn olan en y\u00fcksek hassasiyete, \u00e7\u00f6z\u00fcn\u00fcrl\u00fc\u011fe sahip de\u011fildi. Fakat, \u00f6zellikle de maliyetinin azalmas\u0131yla birlikte, bu tarz haritalama i\u015flemlerinin DNA-hasarlar\u0131n\u0131n hastal\u0131klara ve \u00f6l\u00fcme nas\u0131l sebep oldu\u011funu anlamam\u0131za yard\u0131mc\u0131 olacak rutin i\u015flemler haline gelece\u011fine dair bir ipucu veriyor.<\/p>\n<p>Bu haritaland\u0131rma tekni\u011fi pek \u00e7ok soruya cevap bulunmas\u0131nda yard\u0131mc\u0131 olabilir, \u00f6rne\u011fin:<\/p>\n<ul>\n<li>Bir insanda n\u00fckleotid eksizyon onar\u0131m\u0131 kapasitesine kar\u015f\u0131 koyabilmek i\u00e7in belli bir toksinin ne kadar y\u00fcksek bir dozda bulunmas\u0131 gerekir?<\/li>\n<li>Hangi genlerdeki hangi varyasyonlar insanlar\u0131n daha \u00e7ok veya daha az DNA onar\u0131m\u0131 kapasitesine sahip olmas\u0131na sebep oluyor?<\/li>\n<li>Genomda ba\u015far\u0131l\u0131 onar\u0131m\u0131n ger\u00e7ekle\u015fmesi ihtimalinin daha az oldu\u011fu belli noktalar var m\u0131?<\/li>\n<\/ul>\n<p>Ortalama hassasiyete sahip olan bu ilk haritalamayla bile Sancar ve ekibi, BPDE\u2019nin, sitozinlerin (C) yak\u0131n\u0131ndaki guaninleri (G), timin (T) veya adenin (A) yak\u0131n\u0131ndaki guaninlere oranla daha \u00e7ok etkiledi\u011fini g\u00f6stermeyi ba\u015fard\u0131. Bu da g\u00f6steriyor ki genom \u00fczerinde BPDE taraf\u0131ndan etkilenmeye daha yatk\u0131n olan belirli b\u00f6lgeler var.<\/p>\n<p>Li bu bilginin \u00f6nemini \u015fu s\u00f6zlerle vurguluyor: \u201cBu \u00e7ifte standard\u0131n anla\u015f\u0131lmas\u0131, BaP gibi toksinlere maruz kalman\u0131n neden belli genler \u00fczerinde mutasyon olu\u015fmas\u0131na sebep oldu\u011funu daha iyi anlamam\u0131z\u0131 sa\u011flayabilir.\u201d<\/p>\n<p><strong>\u0130leriye bakarken<\/strong><\/p>\n<p>2015 ve 2016\u2019da yay\u0131mlanan \u00e7al\u0131\u015fmalarda, Sancar ve ekip arkada\u015flar\u0131 \u015fu anda kulland\u0131klar\u0131 haritalama tekni\u011finin daha eski bir versiyonunu kullanarak, biri ultraviyole \u0131\u015f\u0131\u011f\u0131 kaynakl\u0131 di\u011feri ise yayg\u0131n kimyasal ila\u00e7 cisplatin kaynakl\u0131 iki farkl\u0131 DNA-kat\u0131m hasar tipini daha haritalad\u0131. Sancar\u2019a g\u00f6re, \u201cBu yeni metot n\u00fckleotid eksizyon onar\u0131m\u0131 mekanizmas\u0131n\u0131 i\u00e7eren her \u00e7e\u015fit DNA hasar\u0131na uygulanabilir.\u201d<\/p>\n<p>Sancar, Li ve ekip arkada\u015flar\u0131 \u015fimdilerde di\u011fer \u00e7evresel toksinlerle ba\u011flant\u0131l\u0131 DNA hasar\u0131 onar\u0131m mekanizmalar\u0131n\u0131 haritalamak i\u00e7in yeni bir y\u00f6ntem kullan\u0131yorlar. S\u0131radaki projeleri, k\u00fcf mantarlar\u0131 taraf\u0131ndan \u00fcretilen ve k\u00f6t\u00fc bir \u015fekilde saklanan f\u0131nd\u0131klarda ve tah\u0131llarda s\u0131k\u00e7a rastlanan aflatoksinler \u00fczerine. Bu toksinler DNA\u2019ya hasar veriyorlar ve geli\u015fmekte olan \u00fclkelerde karaci\u011fer kanserinin en b\u00fcy\u00fck sebeplerinden biriler.<\/p>\n<p>Ara\u015ft\u0131rmac\u0131lar ayr\u0131ca n\u00fckleotid eksizyon onar\u0131m\u0131n\u0131n ger\u00e7ekle\u015fip ger\u00e7ekle\u015fmeyece\u011fini veya nerede ger\u00e7ekle\u015fece\u011fini etkileyen fakt\u00f6rleri ortaya \u00e7\u0131karmaya \u00e7al\u0131\u015f\u0131yor. Bunun i\u00e7in\u00a0 sadece DNA onar\u0131m\u0131 s\u0131ras\u0131nda kesilen hasarl\u0131 par\u00e7alar\u0131 bulmalar\u0131 de\u011fil, b\u00fct\u00fcn genomun \u00fczerinde ger\u00e7ekten hasarl\u0131 olan her noktan\u0131n haritas\u0131n\u0131 \u00e7\u0131karmalar\u0131 gerekiyor.<\/p>\n<p>Bu tarz bir proje \u00fczerinde \u00e7al\u0131\u015f\u0131l\u0131rken, DNA \u00fczerinde ultraviyole \u0131\u015f\u0131nlar\u0131 dolay\u0131s\u0131yla ortaya \u00e7\u0131kan DNA hasar\u0131n\u0131n genom \u00fczerindeki haritas\u0131n\u0131n \u00e7\u0131kar\u0131lmas\u0131n\u0131 \u00e7ok hassas ve y\u00fcksek kaliteyle ba\u015faran bir teknik daha \u00f6nce geli\u015ftirilmi\u015fti. Bu tekni\u011fi DNA onar\u0131m\u0131 haritalanmas\u0131 tekni\u011fiyle birle\u015ftirdiklerinde, DNA hasar\u0131n\u0131n genomda homojen, e\u015fit bir da\u011f\u0131l\u0131m g\u00f6sterdi\u011fini fakat DNA onar\u0131m\u0131 i\u00e7in ayn\u0131 \u015feyin s\u00f6z konusu olmad\u0131\u011f\u0131n\u0131 g\u00f6rd\u00fcler.
