Yani \u201cotofaji\u201d kendi kendini yemek demek. Bu kavram asl\u0131nda 1960\u2019larda tan\u0131mlanm\u0131\u015ft\u0131. Ara\u015ft\u0131rmac\u0131lar o d\u00f6nemde, h\u00fccrenin kendi i\u00e7eri\u011fini y\u0131k\u0131ma u\u011fratarak h\u00fccre zar\u0131 arac\u0131l\u0131\u011f\u0131yla bu materyali \u201clizozom\u201d ad\u0131 verilen bir \u201cgerid\u00f6n\u00fc\u015f\u00fcm\u201d b\u00f6l\u00fcm\u00fcne ta\u015f\u0131d\u0131\u011f\u0131n\u0131 ve orada bozundurdu\u011funu g\u00f6stermi\u015flerdi. \u00c7al\u0131\u015fma s\u0131ras\u0131ndaki baz\u0131 zorluklar 1990\u2019lara kadar konunun detaylar\u0131na inilmesini geciktirmi\u015fti. 1990\u2019larda Yoshinori Ohsumi ve ekibinin ekmek mayas\u0131 kullanarak yapt\u0131klar\u0131 bir dizi parlak \u00e7al\u0131\u015fma sonucunda otofaji s\u00fcrecinde yer alan temel genler belirlenebildi. Bu da a\u00e7l\u0131\u011fa uyum sa\u011flama veya enfeksiyonlara yan\u0131t verme gibi \u00e7e\u015fitli fizyolojik s\u00fcre\u00e7lerimizde otofajinin temelde nas\u0131l bir \u00f6nem ta\u015f\u0131yabilece\u011fini anlamam\u0131z\u0131 sa\u011flayacakt\u0131. Otofaji genlerinin mutasyonu hastal\u0131klara sebep oluyor olabilir. Sonu\u00e7ta di\u011fer lizozomlarla birle\u015fir.<\/figcaption><\/figure>\n<p><strong>Bozulma ya\u015fayan t\u00fcm h\u00fccrelerin esas fonksiyonu<\/strong><\/p>\n<p>1950\u2019 lerin ortalar\u0131nda biliminsanlar\u0131, i\u00e7inde protein, karbonhidrat ve ya\u011flar\u0131 sindiren enzimlerin bulundu\u011fu ve \u201clizozom\u201d ad\u0131 verilen yeni bir h\u00fccre organeli tan\u0131mlad\u0131. Bu \u00f6zel organel, h\u00fccresel kurucu unsurlar\u0131n bozunmas\u0131nda adeta bir istasyon gibi \u00e7al\u0131\u015f\u0131yordu. Bel\u00e7ikal\u0131 biliminsan\u0131 Christian de Duve, lizozomun ke\u015ffiyle 1974 Nobel T\u0131p\/Fizyoloji \u00d6d\u00fcl\u00fc\u2019n\u00fc alm\u0131\u015ft\u0131. 1960\u2019larda ise yeni ke\u015fifler, \u00e7ok b\u00fcy\u00fck miktarda h\u00fccre i\u00e7eri\u011finin, hatta t\u00fcm organellerin zaman zaman lizozomun i\u00e7inde olabildi\u011fini g\u00f6stermi\u015fti. H\u00fccrelerin bu y\u00fczden, lizozoma b\u00fcy\u00fccek kargo ta\u015f\u0131mac\u0131l\u0131\u011f\u0131 yapma konusunda stratejileri varm\u0131\u015f gibi g\u00f6r\u00fcn\u00fcyordu. Daha fazla biyokimyasal ve mikroskobik analiz sonucu, bu \u201cta\u015f\u0131mac\u0131l\u0131\u011f\u0131\u201d sa\u011flayan yeni bir kesecik oldu\u011fu g\u00f6sterildi (\u015eekil 1). Lizozomun k\u00e2\u015fifi Christian de Devu, bu s\u00fcreci de tan\u0131mlayarak \u201ckendi kendini yeme\u201d i\u015fleyi\u015fine \u201cotofaji\u201d, yeni keseci\u011fe ise \u201cotofagozom\u201d ad\u0131n\u0131 verdi.<\/p>\n<p>1970\u2019ler ve 80\u2019lerde ise ara\u015ft\u0131rmac\u0131lar daha \u00e7ok, \u201cproteazom\u201d ad\u0131n\u0131 verdikleri, hasar g\u00f6rm\u00fc\u015f veya gereksinim olmayan h\u00fccresel proteinlerin par\u00e7alanmas\u0131nda i\u015flev g\u00f6ren enzimatik kompleksler birli\u011finin ke\u015ffine \u00e7al\u0131\u015ft\u0131. Sonu\u00e7ta \u00f6karyot h\u00fccrelerde yayg\u0131n olarak bulunan, h\u00fccre proteinlerinin proteazlarla par\u00e7alanabilmesi i\u00e7in par\u00e7alanacak proteine kovalent ba\u011flarla ba\u011flanan, enzim \u00f6zelli\u011fi bulunmayan, s\u0131cakl\u0131\u011fa dayan\u0131kl\u0131, 76 aminoasitlik k\u00fc\u00e7\u00fck bir protein olan ubikitin arac\u0131l\u0131\u011f\u0131yla protein par\u00e7alanmas\u0131 yola\u011f\u0131n\u0131 ke\u015ffeden Aaron Ciechanover, Avram Heershko ve Irwin Rose\u2019dan ou\u015fan ayn\u0131 ekip 2004 y\u0131l\u0131nda Nobel Kimya \u00d6d\u00fcl\u00fc\u2019ne lay\u0131k g\u00f6r\u00fcld\u00fc. Proteazom, proteinlerin tek tek y\u0131k\u0131m\u0131nda etkiliydi, ancak bu mekanizma, h\u00fccrenin daha b\u00fcy\u00fck protein komplekslerinden ve haraplanm\u0131\u015f organellerden nas\u0131l kurtulabildi\u011fini a\u00e7\u0131klayam\u0131yordu. Bir sonraki ad\u0131m olarak, sa\u011f resimde g\u00f6r\u00fcld\u00fc\u011f\u00fc gibi binlerce maya h\u00fccresiyle \u00e7al\u0131\u015f\u0131larak otofajinin temel s\u00fcrecine dahil olan en az 15 gen belirlendi.<\/figcaption><\/figure>\n<p><strong>\u00c7\u0131\u011f\u0131r a\u00e7an deney<\/strong><\/p>\n<p>Yoshinori Ohsumi, \u00e7e\u015fitli ara\u015ft\u0131rmalarda aktifti, ancak 1988\u2019de kendi laboratuvar\u0131n\u0131 kurdu\u011funda, enerjisini, protein y\u0131k\u0131m\u0131nda rol oynayan bir organel olan ve insanda lizozomla benzerlik g\u00f6steren \u201cvakuol\u201d (Baz\u0131 tekh\u00fccreli hayvanlar\u0131n d\u0131\u015f\u0131nda hayvanlarda bulunmayan bir organeldir. T\u00fcm ilkel ve y\u00fcksek yap\u0131l\u0131 bitkilerde ve mantar h\u00fccrelerinde vard\u0131r. Vakuol \u2013koful- s\u0131v\u0131s\u0131 i\u00e7inde, h\u00fccrenin ait oldu\u011fu doku \u00e7e\u015fidine ba\u011fl\u0131 olarak organik maddeler ve asitler, \u00e7e\u015fitli renk maddeleri, tuzlar, karbonhidratlar, ya\u011flar, proteinler, m\u00fcsilaj, kau\u00e7uk ve tanen maddeler bulunabilir) i\u00e7inde olup bitene yo\u011funla\u015ft\u0131rd\u0131. Ekmek mayas\u0131 h\u00fccreleri g\u00f6receli olarak \u00fczerinde \u00e7al\u0131\u015fmas\u0131 kolay, bu y\u00fczden de insan \u00e7al\u0131\u015fmalar\u0131nda model olarak s\u0131k\u00e7a kullan\u0131labilen h\u00fccrelerdir. \u00d6zellikle de, \u00f6nemli kompleks h\u00fccresel yolaklarda i\u015fleyen genleri belirlemeye yard\u0131mc\u0131 olurlar. Ancak Ohsumi b\u00fcy\u00fck bir engelle kar\u015f\u0131la\u015fm\u0131\u015ft\u0131. Bu h\u00fccreler k\u00fc\u00e7\u00fckt\u00fc ve ba\u015flang\u0131\u00e7 yap\u0131lar\u0131 mikroskop alt\u0131nda kolayl\u0131kla se\u00e7ilemiyorlard\u0131. Bu nedenle de otofajinin var olup olmad\u0131\u011f\u0131 kesin olarak s\u00f6ylenemezdi. Ohsumi bu sebeple, e\u011fer otofajinin aktif s\u00fcrecinde y\u0131k\u0131m i\u015flemini bozarsa, otofagozomlar\u0131n vakuol i\u00e7inde birikece\u011fini ve mikroskop alt\u0131nda g\u00f6r\u00fcnecek kadar bir kitle olu\u015fturabilece\u011fini \u00f6ng\u00f6rd\u00fc. O bu y\u00fczden a\u00e7 ve mutasyona u\u011fram\u0131\u015f maya h\u00fccrelerinde vakuolar y\u0131k\u0131m enzimlerini eksik b\u0131rakarak otofajiyi kendili\u011finden tetikledi. Sonu\u00e7lar \u00e7arp\u0131c\u0131yd\u0131. Saatler i\u00e7inde vakuoller, y\u0131k\u0131ma u\u011framam\u0131\u015f \u00e7ok say\u0131da k\u00fc\u00e7\u00fck kesecikle dolmu\u015ftu (\u015eekil 2). Bu kesecikler otofagozomlard\u0131 ve Ohsumi, deneyiyle maya h\u00fccrelerinde de \u201cotofaji\u201d oldu\u011funu g\u00f6stermi\u015fti. Ancak daha da \u00f6nemlisi bu deneyle, s\u00fcre\u00e7te yer alan genleri belirlemi\u015fti. Bu birikme, otofajide \u00f6nemli olan genlerin i\u015flevsizli\u011fi durumunda g\u00f6zlemlenmiyordu. Ohsumi, maya h\u00fccrelerini kimyasallara maruz b\u0131rakarak mutasyonu pek \u00e7ok gende rasgele olu\u015fturdu\u011funda, otofajiyi tetikledi\u011fini g\u00f6rd\u00fc. Stratejisi i\u015fe yaram\u0131\u015ft\u0131. Bir y\u0131l i\u00e7ersinde Ohsumi, otofaji s\u00fcrecinde temel rol oynayan ilk genleri tan\u0131mlam\u0131\u015ft\u0131. \u0130lerleyen \u00e7al\u0131\u015fmalarda ise bu genler taraf\u0131ndan kodlanan proteinleri i\u015flevsel olarak karakterize edebildi. Sonu\u00e7lar, otofajinin, ard\u0131\u015f\u0131k protein ve protein kompleksleri taraf\u0131ndan kontrol edildi\u011fini, otofajinin her ba\u015flama ve d\u00fczenlenme a\u015famas\u0131n\u0131n belirgin bir gen taraf\u0131ndan d\u00fczenlendi\u011fini g\u00f6stermi\u015fti (\u015eekil 3).<\/p>\n<p><strong>Otofaji: H\u00fccrelerimizin ka\u00e7\u0131n\u0131lmaz\u0131<\/strong><\/p>\n<p>Maya h\u00fccrelerinde otofaji d\u00fczene\u011finin belirlenmesinden sonra geriye kritik bir soru kalm\u0131\u015ft\u0131. Di\u011fer organzimalarda da bu s\u00fcreci kontrol eden benzer bir mekanizma var m\u0131? Yak\u0131n zamanda bizim h\u00fccrelerimizde de neredeyse tamamen ayn\u0131 bir mekanizma oldu\u011fu a\u00e7\u0131kl\u0131k kazand\u0131. \u015eimdi ise bunun insan bedenindeki \u00f6nemini ke\u015ffedecek ara\u015ft\u0131rma gere\u00e7lerine ihtiyac\u0131m\u0131z var.<\/p>\n<p>Ohsumi ve onu izleyen di\u011fer ara\u015ft\u0131rmac\u0131lar sayesinde bug\u00fcn, h\u00fccresel i\u00e7eriklerin bir kendini yok etme-gerid\u00f6n\u00fc\u015f\u00fcm s\u00fcrecine gerek duyduklar\u0131n\u0131 ve bunu otofaji y\u00f6ntemiyle yapabildiklerini \u00f6\u011frendik. Otofaji, h\u0131zl\u0131 bir \u015fekilde enerji sa\u011flay\u0131p h\u00fccresel i\u00e7eriklerin yenilenmesi i\u00e7in gereken ortam\u0131 yaratabiliyor. Tam da bu sayede, h\u00fccrenin a\u00e7l\u0131\u011fa ve di\u011fer stress fakt\u00f6rlerine yan\u0131t verebilmesinde esas g\u00f6revi g\u00f6r\u00fcyor. Enfeksiyondan sonra otofaji, istilac\u0131 h\u00fccre i\u00e7i bakterileri ve vir\u00fcsleri saf d\u0131\u015f\u0131 edebiliyor. Yine otofaji, embriyonun geli\u015fimi ve h\u00fccre farkl\u0131l\u0131a\u015fmas\u0131na yard\u0131mc\u0131 olabiliyor. H\u00fccreler otofajiyi ayn\u0131 zamanda, hasar g\u00f6rm\u00fc\u015f protein ve organellerin at\u0131lmas\u0131nda da kullanabiliyor. Bu da ya\u015flanma s\u00fcrecinin olas\u0131 olumsuz sonu\u00e7lar\u0131n\u0131 \u00f6nlemede kritik bir \u00f6nem ta\u015f\u0131yabiliyor.<\/p>\n<p>Bozulmu\u015f otofaji, Parkinson hastal\u0131\u011f\u0131, tip 2 diyabet ve ya\u015fl\u0131l\u0131kla ba\u011flant\u0131l\u0131 di\u011fer hastal\u0131klarda da rol oynuyor. Otofaji genlerinin mutasyonu genetk hastal\u0131klara sebep olabiliyor. Yine bu bozulma kanserle de ili\u015fkili gibi g\u00f6r\u00fcn\u00fcyor. \u0130lerlemi\u015f \u00e7al\u0131\u015fmalar \u015fimdilerde, ciddi hastal\u0131klarda do\u011frudan otofajiyi hedefleyen ila\u00e7lar geli\u015ftirme yolunda.<\/p>\n<p>Otofaji yakla\u015f\u0131k 50 y\u0131ld\u0131r biliniyor. Ancak fizyoloji ve t\u0131ptaki temel \u00f6nemi 90\u2019larda Yoshinori Ohsumi\u2019nin paradigma de\u011fi\u015ftiren ara\u015ft\u0131rmalar\u0131ndan sonra fark edildi.
