Bu ikinci vir\u00fcs\u00fcn bir insan grip vir\u00fcs\u00fc olmas\u0131 durumunda ise, ortaya insandan insan kolayl\u0131kla bula\u015fabilen bir vir\u00fcs\u00fcn \u00e7\u0131kma olas\u0131l\u0131\u011f\u0131 var. Vir\u00fcs, \u015fimdiden olduk\u00e7a yol alm\u0131\u015f durumda. H5N1, evrim basamaklar\u0131n\u0131 h\u0131zla t\u0131rman\u0131yor. <\/em><\/h4>\n<p>B\u00fcy\u00fck usta Sait Faik\u2019in \u201cSon Ku\u015flar\u201d isimli enfes \u00f6yk\u00fcs\u00fc bir s\u00fcredir akl\u0131mdan \u00e7\u0131km\u0131yor. Yaz gelince leyleklere neler yapacaklar acaba?<\/p>\n<p><strong><em>\u00d6yk\u00fc 1918&#8217;de ba\u015flad\u0131<\/em><\/strong><\/p>\n<p>B\u00fct\u00fcn bunlar, hikayenin bilebildi\u011fimiz k\u0131sm\u0131yla, yakla\u015f\u0131k 87 y\u0131l \u00f6nce ku\u015flara ait bir grip vir\u00fcs\u00fcn\u00fcn insanlar\u0131 infekte etmesi ile ba\u015flad\u0131. 1918 grip salg\u0131n\u0131 en b\u00fcy\u00fck insan kayb\u0131na \u0130spanya\u2019da neden oldu\u011fu i\u00e7in, &#8220;\u0130spanyol Gribi&#8221; olarak tarihe ge\u00e7ti. K\u0131sa bir s\u00fcre \u00f6nce, molek\u00fcler biyologlar\u0131n \u00e7abalar\u0131 ile, \u0130spanyol gribine neden olan vir\u00fcs\u00fcn asl\u0131nda bir ku\u015f gribi vir\u00fcs\u00fcnden evrimle\u015fti\u011fi anla\u015f\u0131ld\u0131. Di\u011fer bir deyi\u015fle, ku\u015flara \u00f6zg\u00fc olan bir grip vir\u00fcs\u00fc, insana bula\u015fma ve onu hasta etme yetisi kazanm\u0131\u015ft\u0131. Farkl\u0131 t\u00fcrlere ait vir\u00fcslerin normalde, kendi orijinal konaklar\u0131 d\u0131\u015f\u0131ndaki canl\u0131lara bula\u015fma ihtimali olduk\u00e7a d\u00fc\u015f\u00fck de olsa bu olas\u0131l\u0131k her zaman vard\u0131r. \u00c7\u00fcnk\u00fc vir\u00fcsler, di\u011fer t\u00fcm canl\u0131lar gibi, genetik yani kal\u0131tsal maddesinde de\u011fi\u015fiklik ge\u00e7irmeye a\u00e7\u0131k canl\u0131lard\u0131r. D\u00fcnyam\u0131zdaki b\u00fcy\u00fck canl\u0131 \u00e7e\u015fitli\u011finin kayna\u011f\u0131 bu de\u011fi\u015fmelerdir. Biyolojik evrim olarak tan\u0131mlanan bu s\u00fcre\u00e7 sonunda d\u00fcnyam\u0131zda daha \u00f6nce varolmayan yeni organizmalar\u0131n ortaya \u00e7\u0131kmas\u0131 m\u00fcmk\u00fcn olmu\u015ftur. \u00dcreme ko\u015fullar\u0131, canl\u0131lar\u0131n kal\u0131tsal yap\u0131lar\u0131nda \u00e7e\u015fitli yollarla meydana gelen bu de\u011fi\u015fmelerden hangilerinin bir sonraki nesle iletilece\u011finde \u00e7ok belirleyicidir.<\/p>\n<p><strong><em>Vir\u00fcs, girebilece\u011fi h\u00fccre ar\u0131yor<\/em><\/strong><\/p>\n<p>\u015eu anda ku\u015f gribi vir\u00fcs\u00fcyle ilgili ya\u015fananlar, tamamen bu vir\u00fcs\u00fcn evrimiyle ilgilidir. Ku\u015f gribi hastal\u0131\u011f\u0131na neden olan vir\u00fcsler, influenza A olarak tan\u0131mlanan bir gurubun \u00fcyesidir. 1918 y\u0131l\u0131nda \u0130spanyol gribine neden olan vir\u00fcs H1N1 olarak tan\u0131mlanm\u0131\u015ft\u0131. \u201cH\u201d harfi bu vir\u00fcs\u00fcn y\u00fczeyini kaplayan koruyucu k\u0131l\u0131f\u0131n d\u00f6rt protein molek\u00fcl\u00fcnden biri olan, \u201chemagl\u00fctinin-HA\u201d proteinini temsil etmektedir. HA\u2019n\u0131n g\u00f6revi, vir\u00fcs\u00fcn enfekte edece\u011fi h\u00fccrenin y\u00fczeyine ba\u011flanmas\u0131n\u0131 sa\u011flamakt\u0131r. Vir\u00fcsler kendi kendini \u00e7o\u011faltmalar\u0131n\u0131 sa\u011flayacak mekanizman\u0131n elemanlar\u0131n\u0131n b\u00fcy\u00fck bir k\u0131sm\u0131ndan yoksun olduklar\u0131ndan, \u00fcremek i\u00e7in, mutlaka sistemlerini kullanabilecekleri bir konak h\u00fccreye gereksinim duyarlar. Vir\u00fcsler bu ama\u00e7la, tek h\u00fccreli bakteriler dahil, her t\u00fcrl\u00fc canl\u0131ya ait h\u00fccreleri kullanabiliyorlar. \u0130\u015fte viral enfeksiyonunun bu a\u015famas\u0131 \u00e7ok \u00f6nemlidir, \u00e7\u00fcnk\u00fc vir\u00fcs\u00fcn bu y\u00fczey proteini konak h\u00fccrenin y\u00fczeyindeki al\u0131c\u0131 molek\u00fclleri tan\u0131yamazsa, vir\u00fcs\u00fcn o h\u00fccrenin i\u00e7ine girip, kendini \u00e7o\u011faltmas\u0131 olas\u0131 de\u011fildir. 566 amino asitten olu\u015fan bu b\u00fcy\u00fck protein molek\u00fcl\u00fcn\u00fc kodlayan gende meydana gelen mutasyonlar sayesinde, ku\u015f gribi vir\u00fcs\u00fcn\u00fcn di\u011fer t\u00fcrlere ait h\u00fccrelere ba\u011flanmas\u0131 m\u00fcmk\u00fcn olabilmektedir.<\/p>\n<figure id=\"attachment_244\" aria-describedby=\"caption-attachment-244\" style=\"width: 476px\" class=\"wp-caption aligncenter\"><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-244 size-full\" src=\"https:\/\/bilimvegelecek.com.tr\/wp-content\/uploads\/2017\/04\/2-1.jpg\" alt=\"H5N1'in yap\u0131s\u0131n\u0131n \u015femayla g\u00f6sterili\u015fi. \" width=\"476\" height=\"315\" srcset=\"https:\/\/bilimvegelecek.com.tr\/wp-content\/uploads\/2017\/04\/2-1.jpg 476w, https:\/\/bilimvegelecek.com.tr\/wp-content\/uploads\/2017\/04\/2-1-300x199.jpg 300w\" sizes=\"auto, (max-width: 476px) 100vw, 476px\" \/><figcaption id=\"caption-attachment-244\" class=\"wp-caption-text\">H5N1&#8217;in yap\u0131s\u0131n\u0131n \u015femayla g\u00f6sterili\u015fi.<\/figcaption><\/figure>\n<p>H1N1 vir\u00fcs\u00fcn\u00fcn bu \u015fekilde tan\u0131mlanmas\u0131n\u0131n nedeni HA proteininin 16 alt tipinden (H1-H16) ilkini i\u00e7eriyor olmas\u0131d\u0131r. \u00d6rne\u011fin, \u0130spanyol Gribinden sonra Asya Gribine neden olan vir\u00fcs, influenza A\u2019n\u0131n H2N2 alt tipiydi. Yani bunda HA proteinin di\u011fer bir tipi bulundu\u011fundan, H2 olarak adland\u0131r\u0131lm\u0131\u015ft\u0131. 1997 y\u0131l\u0131nda Hong Kong Gribine neden olan ve \u00fclkemizde de g\u00f6r\u00fclen etmen, ku\u015f gribi vir\u00fcs\u00fcn\u00fcn H5N1 alt tipiydi. HA proteininin H5 tipi olduk\u00e7a tehlikeli bir molek\u00fcld\u00fcr. H1 molek\u00fcl\u00fc 1918 y\u0131l\u0131nda ku\u015f gribi vir\u00fcs\u00fcn\u00fcn insana ge\u00e7mesine \u00e7ok yard\u0131mc\u0131 olmu\u015ftu, fakat vir\u00fcs\u00fcn sadece akci\u011fer h\u00fccrelerine tutunabilmesini sa\u011flayabilmi\u015fti. Hemagl\u00fctinin y\u00fczey proteininin H5 tipi ise ge\u00e7irdi\u011fi mutasyonlar sonucu, ku\u015f gribi vir\u00fcs\u00fcn\u00fcn akci\u011fer d\u0131\u015f\u0131nda insan\u0131n di\u011fer doku h\u00fccrelerine de ba\u011flanabilmesinin yolunu a\u00e7m\u0131\u015ft\u0131r.<\/p>\n<p><strong><em>H5N1 neden korku sal\u0131yor?<\/em><\/strong><\/p>\n<p>\u015eu anda herkesin merak etti\u011fi \u015fey, H5N1\u2019in insandan insana ge\u00e7mesinin m\u00fcmk\u00fcn olup olamayaca\u011f\u0131. Bunun i\u00e7in H5 molek\u00fcl\u00fcnde yeni birtak\u0131m k\u00fc\u00e7\u00fck mutasyonlar\u0131n olmas\u0131 gerekiyor.<\/p>\n<p>Ku\u015f gribi vir\u00fcs\u00fcn\u00fcn y\u00fczeyini \u00f6rten d\u00f6rt proteinden ikincisi, \u201cNA\u201d k\u0131saltmas\u0131 ile tan\u0131mlanan, n\u00f6raminidaz enzimidir. Bu 454 amino asitten olu\u015fan bir proteindir. Bu proteinin dokuz alt tipi (N1-N9) vard\u0131r. \u00d6rne\u011fin \u0130spanyol Gribi etmeninde bu proteinin N1 tipi bulunurken, Asya Gribine neden olanda N2 tipi yer al\u0131yordu. Bu proteinin g\u00f6revi konak h\u00fccre i\u00e7inde \u00e7o\u011falan vir\u00fcslerin oradan d\u0131\u015far\u0131 \u00e7\u0131kmas\u0131n\u0131 sa\u011flamakt\u0131r. Bu sayede vir\u00fcslerin yay\u0131lmas\u0131 ger\u00e7ekle\u015fmektedir. Ku\u015f gribi vir\u00fcs\u00fcn\u00fcn insanda ciddi bir salg\u0131na neden olabilmesi i\u00e7in, HA yan\u0131nda, bu molek\u00fclde de baz\u0131 k\u00fc\u00e7\u00fck de\u011fi\u015fikliklerin meydana gelmesi gereklidir. \u00d6rne\u011fin ku\u015f gribi vir\u00fcs\u00fcn\u00fcn HA\u2019s\u0131nda meydana gelebilecek tek bir amino asit de\u011fi\u015fikli\u011finin bile bu molek\u00fcl\u00fcn, insan grip vir\u00fcslerinin HA molek\u00fcllerinin becerisini kazanabilmesinde yeterli oldu\u011fu saptanm\u0131\u015ft\u0131r. NA proteini de insan grip vir\u00fcslerindeki NA\u2019ya yak\u0131n bir yap\u0131sal \u00f6zelli\u011fe sahiptir. Yani onlara d\u00f6n\u00fc\u015fmesi i\u00e7in az say\u0131daki mutasyon yeterli olabilecektir. Buradan H5N1\u2019in neden bu kadar korku sald\u0131\u011f\u0131 anla\u015f\u0131labilir.<\/p>\n<p><strong><em>Vir\u00fcs, evrim basamaklar\u0131n\u0131 h\u0131zla t\u0131rman\u0131yor<\/em><\/strong><\/p>\n<p>\u00dc\u00e7\u00fcnc\u00fc ve d\u00f6rd\u00fcnc\u00fc proteinler M1 ve M2 k\u0131saltmalar\u0131 ile g\u00f6sterilmektedir. M1&#8217;in g\u00f6revi, ku\u015f gribi vir\u00fcs\u00fcn\u00fcn kal\u0131tsal maddesi olan tek iplik\u00e7ikli RNA molek\u00fcl\u00fcne ba\u011flanarak onu korumakt\u0131r. M2 proteini ise, vir\u00fcs, h\u00fccre i\u00e7ine girdikten sonra, RNA molek\u00fcl\u00fcn\u00fcn \u00fczerindeki k\u0131l\u0131f\u0131n a\u00e7\u0131larak onun \u00e7\u0131plak hale getirilmesini sa\u011flar. Bu genomda vir\u00fcse ait on gen yer al\u0131r. Bu genlerden d\u00f6rd\u00fcn\u00fcn \u00fcr\u00fcn\u00fc olan proteinlerden daha \u00f6nce s\u00f6z edilmi\u015fti. Geri kalan alt\u0131 proteinden \u00fc\u00e7\u00fc bir enzimin olu\u015fumuna kat\u0131lmaktad\u0131r.<\/p>\n<p>Vir\u00fcs RNA\u2019s\u0131n\u0131 konak h\u00fccre i\u00e7inde \u00e7o\u011faltmak i\u00e7in \u201cRNA polimeraz\u201d isimli bir enzime gereksinim vard\u0131r. Bu enzim k\u0131saca PB2, PB1 ve PA olarak adland\u0131r\u0131lan \u00fc\u00e7 protein molek\u00fcl\u00fcn\u00fcn birle\u015fmesinden olu\u015fur. Bunlar 700\u2019\u00fcn \u00fczerinde amino asit i\u00e7eren b\u00fcy\u00fck proteinlerdir. Kal\u0131tsal madde sentezlenmesi i\u015finde RNA polimeraz enzimine yard\u0131m eden bir protein daha bulunur. K\u0131saca \u201cNP\u201d olarak adland\u0131r\u0131lan bu molek\u00fcl\u00fcn ad\u0131 nukleoprotein\u2019dir. \u00d6zellikle ilk \u00fc\u00e7 proteinde meydana gelen mutasyonlar \u00e7ok \u00f6nemlidir \u00e7\u00fcnk\u00fc bir ku\u015f gribi vir\u00fcs\u00fc olan 1918 \u0130spanyol Gribi vir\u00fcs\u00fc ile insan influenza vir\u00fcsleri aras\u0131ndaki amino asit fark\u0131n\u0131n 10 amino asite kadar d\u00fc\u015ft\u00fc\u011f\u00fc saptanm\u0131\u015ft\u0131r. Di\u011fer bir ifade ile, bir ku\u015f gribi vir\u00fcs\u00fcne ait RNA polimeraz enziminin, bir insan grip vir\u00fcs\u00fcn\u00fcn RNA polimeraz\u0131na d\u00f6n\u00fc\u015fmesi i\u00e7in gereken amino asit de\u011fi\u015fikli\u011fi say\u0131s\u0131 sadece 10\u2019dur. Ara\u015ft\u0131rmac\u0131lar bu d\u00f6n\u00fc\u015f\u00fcm\u00fc sa\u011flayacak mutasyonlar\u0131n meydana gelmesi i\u00e7in \u00e7ok uzun bir s\u00fcre beklenmeyece\u011fi g\u00f6r\u00fc\u015f\u00fcndeler. \u00c7\u00fcnk\u00fc H5N1\u2019in insan ve ku\u015flardan izole edilen \u00e7e\u015fitli bireylerinde bu amino asit mutasyonlar\u0131ndan yedi tanesine halihaz\u0131rda rastlan\u0131lm\u0131\u015ft\u0131r. Yani evrimle\u015fmesi son y\u0131llarda h\u0131zlanan H5N1\u2019in a\u015fmas\u0131 gereken engelin \u00e7ok fazla olmad\u0131\u011f\u0131 g\u00f6r\u00fclmekte.<\/p>\n<p>Viral genler taraf\u0131ndan kodlanan son iki protein NS1 ve NEP\u2019tir. NS1 \u201canti interferon protein\u201ddir. G\u00f6revlerinden biri vir\u00fcs\u00fcn konak olarak yerle\u015fece\u011fi organizman\u0131n, vir\u00fcslere kar\u015f\u0131 kulland\u0131\u011f\u0131 \u00f6zel bir savunma sisteminin etkisiz hale getirilmesidir. NS1 proteininde meydana gelebilecek tek bir amino asit de\u011fi\u015fikli\u011finin bile, molek\u00fcl\u00fcn savunma sistemimizi atlatabilmesi i\u00e7in yeterli oldu\u011fu belirlenmi\u015ftir. G\u00f6r\u00fcld\u00fc\u011f\u00fc gibi H5N1, insan vir\u00fcsleri ile aras\u0131ndaki fark\u0131 olduk\u00e7a kapatm\u0131\u015f durumdad\u0131r. NEP proteinin g\u00f6revi ise sentezlenip bir araya paketlenen viral RNA+protein kompleksinin h\u00fccre d\u0131\u015f\u0131na \u00e7\u0131k\u0131\u015f\u0131na yard\u0131mc\u0131 olmakt\u0131r.<\/p>\n<p>Viral genler taraf\u0131ndan kodlanan bu molek\u00fcller i\u00e7inde antiviral ila\u00e7lar\u0131n hedef molek\u00fcl\u00fc olarak se\u00e7ilenler, HA, NA, M2 yani koruyucu y\u00fczey proteinleridir. Ayn\u0131 \u015fekilde v\u00fccudun savunma molek\u00fclleri olan antikorlar\u0131n hedefi de bu proteinlerdir.
