Bu bildi\u011fimiz, s\u00fcregelen bir durumdur. Bilmedi\u011fimiz \u015fey, bu s\u00fcre\u00e7te var olan \u00fc\u00e7\u00fcnc\u00fc bir unsurdur. Bu unsur; bu s\u00fcrece kendili\u011finden dahil olan, varl\u0131\u011f\u0131 ya\u015famsal say\u0131lan bir otostop\u00e7udur: Transpozonlar!<\/p>\n<p><strong>&#8216;\u0130\u00e7inde ihti\u015fam bar\u0131nd\u0131ran&#8217; bir d\u00fcnyaya yolculuk!<\/strong><\/p>\n<p>&#8220;Transpozon&#8221; veya &#8220;Transposable element&#8221;; genomumuzda gizlenen hareketli par\u00e7ac\u0131klar\u0131n, dolay\u0131s\u0131yla da bahsi ge\u00e7en otostop\u00e7ular\u0131n ad\u0131d\u0131r. Bu DNA sekanslar\u0131 genomun i\u00e7inde hareket edebilmekte ve bu sayede genomunda bulundu\u011fu canl\u0131ya zarar verebilmektedir. Bu duruma en bilinen \u00f6rnek, genomda \u00e7e\u015fitli de\u011fi\u015fimlere neden olmas\u0131 nedeniyle bir\u00e7ok hastal\u0131\u011f\u0131n kap\u0131s\u0131n\u0131 aralamas\u0131 olsa da; y\u0131llarca hemofili ve ba\u015fl\u0131ca kanser t\u00fcrleri ile ili\u015fkilendirilen transpozonlar, asl\u0131nda bizlere &#8220;i\u00e7inde ihti\u015fam bar\u0131nd\u0131ran&#8221; bir d\u00fcnyan\u0131n kap\u0131lar\u0131n\u0131 a\u00e7\u0131yor.<\/p>\n<p>D\u00fcnyan\u0131n en \u00f6nemli sitogenetik\u00e7ilerinden kabul edilen, ayn\u0131 zamanda bir ba\u015fka Evrimsel Mekanizma olan &#8220;Crossing Over&#8221;\u0131n da ke\u015ffini ger\u00e7ekle\u015ftiren Nobel Fizyoloji ve T\u0131p \u00f6d\u00fcl\u00fc sahibi Barbara McClintock taraf\u0131ndan ilk defa deneysel olarak g\u00f6zlenen transpozonlar, hastal\u0131klarla ili\u015fkili olmalar\u0131n\u0131n yan\u0131nda varl\u0131klar\u0131 ve hareketleriyle y\u00fcz binlerce y\u0131ld\u0131r do\u011fan\u0131n en muhte\u015fem ger\u00e7e\u011fini \u015fekillendiriyor: Evrim!<\/p>\n<p>Transpozonlar; genetik materyalde bulunan, zaman zaman ve bir ba\u015fka mekanizmaya ba\u011fl\u0131 olmadan kromozomlar \u00fczerinde bir b\u00f6lgeden ba\u015fka bir b\u00f6lgeye s\u0131\u00e7rayabilen DNA dizilimleridir. Transpozonlar\u0131n varl\u0131\u011f\u0131 ve hareketi ise y\u00fczy\u0131llard\u0131r Evrim&#8217;i \u015fekillendiren en temel mekanizmalardan biri olarak kar\u015f\u0131m\u0131za \u00e7\u0131k\u0131yor. Ancak \u015fimdiye kadar yap\u0131lan \u00e7al\u0131\u015fmalarda, transpozonlar\u0131n bu de\u011fi\u015fimi bir sonraki nesile do\u011fru nas\u0131l aktard\u0131\u011f\u0131 ve bu s\u00fcrecin bir sonraki nesilde nas\u0131l devam etti\u011fi ara\u015ft\u0131r\u0131lmam\u0131\u015ft\u0131. Bilim insanlar\u0131 ilk kez, yeni varl\u0131klara d\u00f6n\u00fc\u015fecek olan embriyolar ile gelece\u011fe &#8220;s\u0131\u00e7ramak&#8221; i\u00e7in hedefe transpoze olan h\u00fccrelerin varl\u0131klar\u0131n\u0131 g\u00f6zlemledi.<\/p>\n<figure id=\"attachment_28333\" aria-describedby=\"caption-attachment-28333\" style=\"width: 300px\" class=\"wp-caption alignright\"><img loading=\"lazy\" decoding=\"async\" class=\"size-medium wp-image-28333\" src=\"https:\/\/bilimvegelecek.com.tr\/wp-content\/uploads\/2018\/08\/foto-11-300x184.jpg\" alt=\"\" width=\"300\" height=\"184\" \/><figcaption id=\"caption-attachment-28333\" class=\"wp-caption-text\">\u00c7al\u0131\u015fmay\u0131 y\u00fcr\u00fcten Zhao Zhang, &#8220;S\u0131\u00e7rama genleri, oositler olarak adland\u0131r\u0131lan geli\u015fmekte olan yumurta h\u00fccrelerinin genomuna tercihen dahil olan istilac\u0131 \u00fcr\u00fcnlerin \u00fcretilmesi i\u00e7in hem\u015fire h\u00fccrelerini kullan\u0131r&#8221; diyor.<\/figcaption><\/figure>\n<p><strong>&#8216;Savunmac\u0131 strateji&#8217;: piRNA&#8217;lar<\/strong><\/p>\n<p>Bu noktada; transpozonlar\u0131n varl\u0131klar\u0131n\u0131 ve hareketlerini anlamak i\u00e7in, transpozonlar\u0131n i\u015flevi ve ili\u015fkileri hakk\u0131nda daha fazla bilgi vermek i\u00e7in hareketli elementlerle ilgili Carnegie Science Center&#8217;dan Zhao Zhang ve ekibi taraf\u0131ndan y\u00fcr\u00fct\u00fclen ara\u015ft\u0131rmaya bakal\u0131m. Teoride, transpozonlar\u0131n v\u00fccutta kontrols\u00fcz bir \u015fekilde \u00e7al\u0131\u015ft\u0131r\u0131lmas\u0131na izin verilmi\u015fse, bunlar sadece \u00f6len genetik hatalarla sonu\u00e7lan\u0131r. Bunu yapmak i\u00e7in, belirli bir piRNA t\u00fcr\u00fcn\u00fc kapatt\u0131lar ve &#8220;atlama h\u00fccreleri&#8221;,\u00a0 genin iki germ h\u00fccresinden (her bir ebeveynden birinden gelen) geli\u015ftik\u00e7e onlar\u0131n nas\u0131l hareket ettiklerini izlediler. Bu sayede, &#8220;Retrotransposonlar&#8221; olarak bilinen baz\u0131 atlama genlerinin, geli\u015fmekte olan yumurtalar i\u00e7in protein ve RNA gibi genetik kaynaklar\u0131 \u00fcreten &#8220;hem\u015fire h\u00fccrelere&#8221; dayanmakta oldu\u011funu ke\u015ffettiler. Cornell \u00dcniversitesi&#8217;nden Molek\u00fcler Biyolog Cedric Feschotte ,ara\u015ft\u0131rman\u0131n i\u00e7inde bulunmamas\u0131na ra\u011fmen &#8220;Bu \u00e7al\u0131\u015fma, transpozonlar\u0131n karma\u015f\u0131k ya\u015fam\u0131n\u0131 ortaya koyuyor&#8221; diyor ve transpozonlar\u0131n g\u00f6z kama\u015ft\u0131r\u0131c\u0131 d\u00fcnyas\u0131n\u0131n ortaya \u00e7\u0131kar\u0131lmas\u0131n\u0131n \u00f6nemini vurguluyor.
