Parkinson hastal\u0131\u011f\u0131 Dopamin kimyasal\u0131n\u0131 \u00fcreten ve beyinde bulunan sinir h\u00fccrelerine sald\u0131r\u0131yor, Dopamin \u00fcretemeyen v\u00fccutta; titremeler, kas sertli\u011fi, denge ve koordinasyon sorunlar\u0131 g\u00f6r\u00fcl\u00fcyor. Dolay\u0131s\u0131yla L-DOPA, Parkinson\u2019a ba\u011fl\u0131 sorunlar\u0131n giderilmesi i\u00e7in beyne \u00f6nc\u00fcl maddelerin ula\u015fmas\u0131n\u0131 sa\u011fl\u0131yor. L-DOPA\u2019n\u0131n 1960\u2019lar\u0131n sonunda piyasaya sunulmas\u0131ndan beri ara\u015ft\u0131rmac\u0131lar, v\u00fccuttaki enzimlerin L-DOPA\u2019y\u0131 ba\u011f\u0131rsaklarda sindirerek ilac\u0131n beyne ula\u015fmas\u0131n\u0131 engelleyebilece\u011finin fark\u0131ndayd\u0131lar. Bunun i\u00e7in ila\u00e7 end\u00fcstrisi istenmeyen L-DOPA metabolizmas\u0131n\u0131 bloke etmek i\u00e7in \u201cKarbidopa\u201d ad\u0131nda yeni bir ila\u00e7 piyasaya s\u00fcrd\u00fc. Bu iki ila\u00e7 birlikte al\u0131nd\u0131\u011f\u0131nda tedavi i\u015fe yar\u0131yormu\u015f gibi g\u00f6r\u00fcn\u00fcyordu.<\/p>\n<p>Manini Redekal, \u201cA\u00e7\u0131klayamad\u0131\u011f\u0131m\u0131z \u00e7ok say\u0131da metabolik olay var ve insanlar aras\u0131nda de\u011fi\u015fkenlik g\u00f6steriyor. Bu de\u011fi\u015fkenlik esas problem. \u0130la\u00e7 baz\u0131 hastalar i\u00e7in daha az etkili olmakla kalm\u0131yor, ayn\u0131 zamanda L-DOPA beyin d\u0131\u015f\u0131nda Dopamin\u2019e d\u00f6n\u00fc\u015ft\u00fc\u011f\u00fcnde bu madde ba\u011f\u0131rsaklarda ciddi s\u0131k\u0131nt\u0131lara ve kalp ritim bozuklu\u011fu gibi yan etkilere neden olabiliyor. \u0130lac\u0131n daha az\u0131 beyine ula\u015ft\u0131\u011f\u0131nda \u00e7o\u011fu zaman hastalara bu hastal\u0131\u011f\u0131n semptomlar\u0131n\u0131 ortadan kald\u0131rmak i\u00e7in ilac\u0131n daha fazlas\u0131 veriliyor ve bu durum da g\u00f6r\u00fclen yan etkileri daha k\u00f6t\u00fc hale getirebiliyor.<\/p>\n<p>Manini Redekal ve ekibi, L-DOPA\u2019n\u0131n etkisinin azalarak beyne ula\u015fmas\u0131nda mikroorganizmalar\u0131n etkisi olabilece\u011finden \u015f\u00fcpheleniyorlard\u0131. L-DOPA ile birlikte al\u0131nan antibiyotiklerin, L-DOPA\u2019n\u0131n etkisini artt\u0131\u011f\u0131n\u0131 g\u00f6steren ara\u015ft\u0131rmalar ile birlikte bilim insanlar\u0131, bakterilerin bu konuda temel su\u00e7lu olabilece\u011fini tahmin ediyorlard\u0131. Ancak hen\u00fcz hi\u00e7bir bilim insan\u0131, bakteri t\u00fcrlerinin \u201csu\u00e7lu tutulabilir\u201d durumda olduklar\u0131n\u0131 ve ila\u00e7lar\u0131n bu mikroorganizmalar taraf\u0131ndan neden ve nas\u0131l t\u00fcketildiklerini ke\u015ffedemedi.<\/p>\n<p>Profes\u00f6r Balskus\u2019un ekibi, bu konuya ili\u015fkin bir \u00e7al\u0131\u015fma ba\u015flatt\u0131lar ve L-DOPA\u2019dan Dopamin\u2019e giden ola\u011fand\u0131\u015f\u0131 kimyasal s\u00fcre\u00e7 onlar\u0131n ilk ipu\u00e7lar\u0131yd\u0131. Bu yola\u011fa g\u00f6z atacak olursak; az say\u0131da bakteriyel enzimin bu d\u00f6n\u00fc\u015f\u00fcm\u00fc ger\u00e7ekle\u015ftirebildi\u011fini g\u00f6rebiliriz. Fakat bu enzimlerin \u00e7o\u011fu L-DOPA\u2019ya olduk\u00e7a benzer bir amino asit olan Tirosin\u2019i ba\u011flamay\u0131 tercih edebiliyor. S\u00fctte ve tur\u015fuda bulunan bir mikroorganizma, hem Tirosin\u2019i hem de L-DOPA\u2019y\u0131 kabul edebiliyor.<\/p>\n<p>\u0130nsan Mikrobiyom Projesi\u2019ni referans alan Manini Redekal ve ekibi, ba\u011f\u0131rsakta ya\u015fayan hangi mikroorganizman\u0131n bu t\u00fcrden enzimleri \u00fcretecek genom yap\u0131s\u0131na sahip olduklar\u0131 \u00fczerine ara\u015ft\u0131rma yapt\u0131lar ve bu bakteriler aras\u0131ndan kriterlerine uyan bir bakteriyi fark ettiler. <em>Enterococcus faecalis <\/em>isimli bakteri, her bir denemede sa\u011flanan b\u00fct\u00fcn L-DOPA maddesini t\u00fcketti. Bu \u00e7al\u0131\u015fma ile birlikte; ara\u015ft\u0131rmac\u0131lardan olu\u015fan ekip, <em>E. faecalis<\/em> ve bu bakteri taraf\u0131ndan \u00fcretilen enzimin L-DOPA metabolizmas\u0131na ba\u011fland\u0131\u011f\u0131n\u0131 kan\u0131tlayan ilk g\u00fc\u00e7l\u00fc delili sunmu\u015f oldu.<\/p>\n<p>Ancak bu duruma ra\u011fmen, ba\u011f\u0131rsaklarda bulunan bir enzim, L-DOPA\u2019y\u0131 Dopamin\u2019e d\u00f6n\u00fc\u015ft\u00fcrebiliyor. Asl\u0131nda bu durum Karbidopa isimli ila\u00e7 taraf\u0131ndan durdurulmas\u0131 gereken bir reaksiyonken, durdurulam\u0131yordu. \u00c7al\u0131\u015fma ekibinin merak etti\u011fi temel noktalardan biri olan Karbidopa\u2019n\u0131n neden etki etmedi\u011fi ve bu ilac\u0131n neden<em> E.faecalis<\/em> taraf\u0131ndan \u00fcretilen enzime ula\u015famad\u0131\u011f\u0131yd\u0131.<\/p>\n<p><strong>\u2018Ke\u015ffedilen molek\u00fcl, sadece elzem olmayan bir enzimi hedef al\u0131yor\u2019<br \/>\n<\/strong>\u0130nsan v\u00fccudunda ve bakterilerde \u00fcretilen baz\u0131 enzimler ayn\u0131 kimyasal reaksiyonu ger\u00e7ekle\u015ftiriyor olmas\u0131na ra\u011fmen, bakteriyel kaynakl\u0131 olan enzim biraz daha farkl\u0131 bir \u00e7al\u0131\u015fma prensibine sahip. Maini Redekal, Karbidopa\u2019n\u0131n mikroorganizmalar\u0131 yok edemedi\u011finden veya h\u00fccreler aras\u0131ndaki yap\u0131sal farkl\u0131l\u0131\u011f\u0131n, Karbidopa\u2019n\u0131n bakteriyel enzimle etkile\u015fime girmesini engelledi\u011finden \u015f\u00fcpheleniyordu. E\u011fer bu \u015f\u00fcphe do\u011fruysa, benzer mikroorganizmalara ve hatta daha kompleks organizmalara kar\u015f\u0131 Karbidopa\u2019n\u0131n etkisiz kald\u0131\u011f\u0131 kan\u0131tlanabilirdi.<\/p>\n<p>Ancak bu durumun temel sebebi, problem olu\u015fturacak \u00f6nemde olmayabilir. Balskus ve ekibi, bakteriyel enzimin etkisini yava\u015flat\u0131c\u0131 bir molek\u00fcl buldular. Konuya ili\u015fkin a\u00e7\u0131klama yapan Maini Redekal, \u201cKe\u015ffedilen molek\u00fcl, istenmeyen bakteriyel metabolizman\u0131n \u00e7al\u0131\u015fmas\u0131n\u0131 bakteriyi \u00f6ld\u00fcrmeden engelliyor ve sadece elzem olmayan bir enzimi hedef al\u0131yor\u201d dedi. Ke\u015ffedilen molek\u00fcl ve bu molek\u00fcle benzer yap\u0131da bulunan bile\u015fiklerin, Parkinson hastal\u0131\u011f\u0131na kar\u015f\u0131 L-DOPA tedavisinin daha etkin bir bi\u00e7imde s\u00fcrd\u00fcr\u00fclmesi i\u00e7in yeni ila\u00e7lar\u0131n geli\u015ftirilmesinde \u00f6n a\u00e7\u0131c\u0131 olaca\u011f\u0131 d\u00fc\u015f\u00fcn\u00fclmekte.<\/p>\n<p>Bu \u00e7al\u0131\u015fmay\u0131 y\u00fcr\u00fcten ekip, bu bile\u015fi\u011fin bulunmas\u0131 ile s\u0131n\u0131rl\u0131 kalabilirdi ancak bunun yerine L-DOPA\u2019n\u0131n mikrobiyal metabolizmas\u0131nda ikinci bir basama\u011f\u0131 daha ke\u015ffetmek i\u00e7in biraz daha ileri d\u00fczeyde \u00e7al\u0131\u015fmalar yapmay\u0131 tercih ettiler. <em>E. faecalis; <\/em>bu ilac\u0131n Dopamin\u2019e d\u00f6n\u00fc\u015f\u00fcm\u00fcn\u00fc sa\u011flad\u0131ktan sonra, ba\u015fka bir organizma da Dopamin\u2019i Meta-Tiramin kimyasal maddesine d\u00f6n\u00fc\u015ft\u00fcr\u00fcyor.<\/p>\n<figure id=\"attachment_35057\" aria-describedby=\"caption-attachment-35057\" style=\"width: 300px\" class=\"wp-caption alignleft\"><img loading=\"lazy\" decoding=\"async\" class=\"size-medium wp-image-35057\" src=\"https:\/\/bilimvegelecek.com.tr\/wp-content\/uploads\/2019\/07\/E.-Faecalis-300x201.jpg\" alt=\"\" width=\"300\" height=\"201\" \/><figcaption id=\"caption-attachment-35057\" class=\"wp-caption-text\">L-DOPA&#8217;n\u0131n emilimini azaltan E. Hangi mikroorganizman\u0131n geli\u015fece\u011fini veya geli\u015fim g\u00f6stermeyece\u011fini anlamak i\u00e7in mikroorganizmalar\u0131n hepsine Dopamin verildi.<\/p>\n<p>Bu oyunun kazanan\u0131 <em>Eggerthella lenta <\/em>isimli bir mikroorganizma oldu. Bu bakterilerin, Dopamin t\u00fcketip Meta-Tiramin\u2019i yan \u00fcr\u00fcn olarak \u00fcrettikleri fark edildi.
