Mitokondrilerin canl\u0131 v\u00fccuduna sa\u011flad\u0131klar\u0131 enerji, h\u00fccrenin di\u011fer organelleri taraf\u0131ndan farkl\u0131 yolaklarda kullan\u0131l\u0131r. B\u00f6ylelikle enerji d\u00f6n\u00fc\u015f\u00fcmlerinin v\u00fccudumuzdaki temel ve ilk \u00f6rnekleri h\u00fccrelerimizde g\u00f6zlenir.<\/p>\n<p>Kendilerine has genetik materyalleri ve \u00e7ift zarl\u0131 dokusu ile mitokondriler, ancak ve ancak \u00f6karyotik olarak tan\u0131mlad\u0131\u011f\u0131m\u0131z canl\u0131lar taraf\u0131ndan ta\u015f\u0131nabilirler. Ya\u015fam a\u011fac\u0131nda arkeleri ve bakterileri d\u0131\u015far\u0131da b\u0131rakan \u00f6karyotlarda bulunan ve enerjimizin temelini olu\u015fturan mitokondrilerin, zarl\u0131 organel ta\u015f\u0131mayan canl\u0131lardaki temsili ise enerji \u00fcretimini sa\u011flayan enzimlerdir. Peki; ya\u015famsal metabolik aktivitelerden g\u00fcnl\u00fck ya\u015fant\u0131m\u0131za kadar her noktada ihtiya\u00e7 duydu\u011fumuz enerji nas\u0131l \u00fcretiliyor? V\u00fccudumuzda ne gibi yolaklar\u0131n etkin \u00e7al\u0131\u015fmas\u0131 ile enerji \u00fcretimi sa\u011flan\u0131yor?<\/p>\n<p>Bu noktada, enerji santrallerimiz olan mitokondrileri i\u00e7eren canl\u0131lar ile i\u00e7ermeyen canl\u0131lar \u00fczerinden bir ayr\u0131m yapmam\u0131z gerekmekte. Mitokondri ta\u015f\u0131yan h\u00fccrelere sahip organizmalar, bu organelde \u00fcretilen kimyasal enerjiyi kullanarak kendi aktivitelerini ger\u00e7ekle\u015ftirebilirler. Mitokondri ta\u015f\u0131mayan organizmalarda farkl\u0131 bir s\u00fcre\u00e7 i\u015fler ancak bu canl\u0131lar da kaynaklar\u0131 farkl\u0131 olsa da temelde ayn\u0131 kimyasal enerjiyi kullan\u0131r. Bu kimyasal enerjinin ad\u0131 ATP\u2019dir. ATP a\u00e7\u0131l\u0131m\u0131 itibariyle Adenozin Tri Fosfat; Adenin n\u00fckleik baz\u0131 ve fosfat molek\u00fcllerinden olu\u015fur.<\/p>\n<p><strong>ATP \u00fcretimine bak\u0131\u015f: Solunum mekanizmas\u0131<br \/>\n<\/strong>Canl\u0131 v\u00fccudunda enerjinin \u00fcretilmesini sa\u011flayan temel mekanizma solunumdur. Solunum, oksijen vas\u0131tas\u0131yla ger\u00e7ekle\u015ftirilebilece\u011fi gibi; oksijensiz olarak da ger\u00e7ekle\u015ftirilebilir. Dolay\u0131s\u0131yla v\u00fccuttaki enerji \u00fcretimi oksijen varl\u0131\u011f\u0131na ba\u011fl\u0131 de\u011fildir. Mitokondrilerde ise bu durum biraz daha farkl\u0131. Mitokondriler, temel olarak oksijenli solunumun ger\u00e7ekle\u015fti\u011fi organellerdir. Oksijenli solunum vas\u0131tas\u0131yla \u00fcretilen enerji miktar\u0131n\u0131n daha fazla olmas\u0131 nedeniyle mitokondrilerin, temel yarg\u0131da oldu\u011fu gibi v\u00fccudumuzun enerji santralleri oldu\u011fu ifade edilebilir. Bu noktada, temel sorumuzu biraz daha geni\u015fletelim ve bu soruya yan\u0131t verelim: V\u00fccudumuz nas\u0131l enerji \u00fcretir?<\/p>\n<p>Bu noktada temel odak, solunum mekanizmas\u0131nda olmal\u0131d\u0131r. Genellikle \u201cGlikoliz\u201di, \u201cEtil Alkol Fermantasyonu\u201d ile \u201cLaktik Asit Fermantasyonu\u201d olarak iki ayr\u0131 t\u00fcrde ger\u00e7ekle\u015fen reaksiyonlar izler. Glikoliz\u2019in ba\u015flang\u0131\u00e7 maddesi Glikoz\u2019dur ve bu olay Glikoz\u2019un, Pir\u00fcvik asit\u2019e kadar y\u0131k\u0131m\u0131 ile \u00f6zetlenebilir. Temel metabolik olaylardan solunumun en temel yola\u011f\u0131 olan Glikoliz, b\u00fct\u00fcn canl\u0131larda g\u00f6zlenir ve Glikoliz\u2019in ger\u00e7ekle\u015febilmesi i\u00e7in gerekli enzimler, b\u00fct\u00fcn canl\u0131larda ayn\u0131d\u0131r. Enzimlerin ve yola\u011f\u0131n genel i\u015fleyi\u015finin b\u00fct\u00fcn canl\u0131larda birebir ayn\u0131 olmas\u0131, evrimsel a\u00e7\u0131dan da \u00f6nem ta\u015f\u0131r.<\/p>\n<p>Metabolik olaylar vas\u0131tas\u0131yla ger\u00e7ekle\u015fen ATP \u00fcretimine \u201cFosforilasyon\u201d ad\u0131 verilir ve bu kavram kendi aras\u0131nda birka\u00e7 ba\u015fl\u0131\u011fa ayr\u0131l\u0131r. Bu solunum t\u00fcr\u00fcnde Glikoliz\u2019i, Krebs d\u00f6ng\u00fcs\u00fc ve ETS yola\u011f\u0131 izler. Krebs ve ETS mekanizmalar\u0131n\u0131n ger\u00e7ekle\u015fti\u011fi yer, h\u00fccrelerimizin temel enerji santralleri olan mitokondrilerdir. Bu noktada belirtmek gerekir ki, ana sorumuzun cevab\u0131 mitokondrilerde!<\/p>\n<p>Mitokondriler, \u00f6karyotik h\u00fccrelerde metabolik enerji \u00fcretiminde rol oynar; Oksidatif Fosforilasyon yoluyla ATP \u00fcretiminden sorumludur ve bu organel, kendine \u00f6zg\u00fc bir genom ile bu genomlar taraf\u0131ndan sentezlenen proteinleri ta\u015f\u0131r. Mitokondri, i\u00e7 ve d\u0131\u015f mitokondriyal membranlar i\u00e7erir. \u0130\u00e7 zar organelin i\u00e7 k\u0131sm\u0131na do\u011fru uzanan \u00e7ok say\u0131da k\u0131vr\u0131ml\u0131 yap\u0131ya sahiptir. Bu k\u0131vr\u0131ml\u0131 yap\u0131lara \u201ckrista\u201d ad\u0131 verilir.<\/p>\n<p>Mitokondrinin i\u00e7 zar\u0131n\u0131n d\u0131\u015far\u0131daki alandan ay\u0131rd\u0131\u011f\u0131 s\u0131v\u0131 b\u00f6lge olan matriks, mitokondri genomunu ve oksidatif fosforilasyonun ger\u00e7ekle\u015fmesi i\u00e7in gereken ana \u00f6\u011feleri ta\u015f\u0131r. Glikoliz\u2019in ger\u00e7ekle\u015fmesinin ard\u0131ndan Aeorobik solunum i\u00e7in yoluna devam eden Pir\u00fcvik asit, \u00e7e\u015fitli kanallar yoluyla mitokondriye ta\u015f\u0131n\u0131r. Burada, Krebs D\u00f6ng\u00fcs\u00fc\u2019ne giri\u015f g\u00f6zlenir; Piruvat, CO2 varl\u0131\u011f\u0131 ile Asetil CoA\u2019ya d\u00f6n\u00fc\u015ft\u00fcr\u00fcl\u00fcr. Bu maddenin \u00fcretimi ile Krebs d\u00f6ng\u00fcs\u00fcne ge\u00e7irilir. Krebs Devri di\u011fer bir ad\u0131yla da Sitrik Asit D\u00f6ng\u00fcs\u00fc, hem karbonhidratlar hem de ya\u011f asitleri i\u00e7in oksidatif par\u00e7alanman\u0131n g\u00f6r\u00fcld\u00fc\u011f\u00fc merkez yolakt\u0131r.<\/p>\n<p>Asetil CoA\u2019n\u0131n varl\u0131\u011f\u0131 ile ba\u015flayan Krebs Devri; NAD+ ve FAD koenzimlerinin, NADH ve FADH2\u2019ye indirgenmesiyle sonu\u00e7lan\u0131r. Burada da \u201cSubstrat D\u00fczeyinde Fosforilasyon\u201d g\u00f6zlenir. Sitrik Asit D\u00f6ng\u00fcs\u00fc ile olu\u015fan NADH ve FADH2\u2019ler bir sonraki a\u015fama i\u00e7in \u201cOksidatif Fosforilasyon\u201dun merkezi olan ETS yola\u011f\u0131na aktar\u0131l\u0131r. Burada NADH ve FADH2\u2019den gelen y\u00fcksek enerjiye sahip elektronlar, zar y\u00fczeyinde bulunan bir dizi ta\u015f\u0131y\u0131c\u0131 \u00fczerinden molek\u00fcler oksijene aktar\u0131l\u0131r. Burada membran boyunca \u00fcretilen enerji potansiyel enerjiye d\u00f6n\u00fc\u015ft\u00fcr\u00fcl\u00fcr ve ATP \u00fcretimi i\u00e7in kullan\u0131l\u0131r. Bu noktada i\u00e7 mitokondriyal zar\u0131n, Krebs\u2019in ard\u0131ndan ge\u00e7ilen ETS yola\u011f\u0131nda da ana b\u00f6lge oldu\u011funu belirtmek gerekir.
