Baz\u0131s\u0131n\u0131n ba\u011f\u0131rsaklar\u0131 poliplerle doluydu. Baz\u0131 mutasyonlar h\u00fccre i\u00e7ine girip kendini \u00e7o\u011faltarak sonu\u00e7lara yol a\u00e7arken, di\u011ferleri genleri aktive ederek ya da onlar\u0131 tamamen etkisizle\u015ftirerek molek\u00fcler de\u011fi\u015fikli\u011fe sebep oluyor.<\/p>\n<p>H\u00fccre i\u00e7i materyalde olu\u015fan rasgele farkl\u0131la\u015fman\u0131n yerle\u015fimi, b\u00fcy\u00fck olas\u0131l\u0131kla \u015fansa ba\u011fl\u0131 olarak belirleniyor. Do\u011fal se\u00e7ilim sadece, ya\u015flanman\u0131n ba\u015flad\u0131\u011f\u0131 40 ya\u015f civar\u0131na kadar etraf\u0131m\u0131zda dolan\u0131r.<\/p>\n<p>Gen ifadelerindeki mutasyon ve dalgalanmalar ba\u015flang\u0131\u00e7ta organizman\u0131n adaptasyonu i\u00e7in bir nimetken, ilerleyen ya\u015flarda olu\u015fturaca\u011f\u0131 birikmelerle \u00e7ok fazla gen ifadesi dalgalanmalar\u0131na yol a\u00e7maya ba\u015flar. Yine Martin\u2019e g\u00f6re bu \u015fans belki de kanser ve di\u011fer dejeneratif beyin hastal\u0131klar\u0131 gibi \u201cgeriatrik hastal\u0131klar\u201da \u00f6nc\u00fcl\u00fck ediyor olabilir.<\/p>\n<p>Ya\u015fam\u0131m\u0131z boyunca \u015fans, ufak ve kazara olu\u015fan mutasyonlar arac\u0131l\u0131\u011f\u0131yla v\u00fccudumuza sokuluverir. Zamanla olu\u015fan b\u00f6l\u00fcnmelerle h\u00fccrelerde v\u00fccutsal mutasyonlar meydana gelir. Her b\u00f6l\u00fcnmede olu\u015fabilecek hatalar, DNA\u2019y\u0131 zor durumda b\u0131rak\u0131r. DNA ile kar\u015f\u0131 kar\u015f\u0131ya gelen bir k\u0131s\u0131m genler normalde bu mutasyonlar\u0131 onarabilir. Fakat yeteri kadar zaman ge\u00e7tikten sonra hata, onar\u0131c\u0131 genlerde de ka\u00e7\u0131n\u0131lmaz olarak g\u00f6zlenecektir. \u015eans\u0131n, riskleri art\u0131rma olas\u0131l\u0131\u011f\u0131 tam olarak var diyemeyiz. Ok, \u015fansl\u0131 bir b\u00f6lgeye isabet ederse, benim gibi 89 y\u0131l ya\u015fayabileceksiniz demektir. \u015eanss\u0131z bir b\u00f6lgeye denk gelirse de, \u00f6len e\u015fim gibi beyinde ba\u015flayan ve sald\u0131rgan karakterli bir kansere maruz kalabilirsiniz. E\u015fim, fiziksel yap\u0131s\u0131 \u00e7ok g\u00fc\u00e7l\u00fc olmas\u0131na ra\u011fmen k\u00f6t\u00fc huylu t\u00fcm\u00f6r olu\u015fumuna yol a\u00e7an bask\u0131n (dominant onkojen) genleri taraf\u0131ndan vurulmu\u015ftu\u201d diyor Martin.<\/p>\n<p>T\u00fcm kanserler \u015fanstan kaynaklanmaz. 2015 Ocak ay\u0131nda biyoistatistik ve kanser genetik ara\u015ft\u0131rmac\u0131lar\u0131n\u0131n yapt\u0131klar\u0131 a\u00e7\u0131klamada, kanserlerin \u00fc\u00e7te birinin kal\u0131tsal defektlere ve g\u00fcne\u015f \u0131\u015f\u0131\u011f\u0131ndan sigaraya uzanan geni\u015f yelpazedeki sald\u0131rgan \u00e7evresel fakt\u00f6rlere ba\u011fl\u0131 olabilece\u011fi belirtilmi\u015fti. Artakalanlar da belki Science dergisinde yay\u0131mland\u0131\u011f\u0131 gibi, \u015fans mutasyonlar\u0131 gibi rasgele i\u00e7sel olaylara ba\u011fl\u0131yd\u0131.<\/p>\n<p>Son zamanlarda yap\u0131lan \u00e7al\u0131\u015fmalarsa, ihmal edilebilir g\u00f6r\u00fcnen rasgele mutasyonlar\u0131n nas\u0131l olup da zamanla birikerek bir sorun olu\u015fturabildi\u011fi \u00fczerine yo\u011funla\u015ft\u0131. Bu mutasyonlar\u0131n baz\u0131lar\u0131 gen ifadelerinde de\u011fi\u015fikli\u011fe sebep olabiliyordu. En geli\u015fkin teknolojiler, h\u00fccre biyologlar\u0131na, v\u00fccut i\u00e7i ortamda bireysel gen ifadelerinde olu\u015fan anl\u0131k farkl\u0131l\u0131klar\u0131 analiz etme \u015fans\u0131 tan\u0131m\u0131\u015ft\u0131. Gen aktivasyonlar\u0131, uyumlu RNA d\u00fczeyleri arac\u0131l\u0131\u011f\u0131yla yorumlanm\u0131\u015ft\u0131. Zira RNA, molek\u00fcler yolakta, gen ve protein aras\u0131ndaki arac\u0131yd\u0131. H. Waddington, h\u00fccreler aras\u0131ndaki azal\u0131p \u00e7o\u011falma fazlal\u0131\u011f\u0131n\u0131n zararl\u0131 olabilece\u011fini ve v\u00fccutta bunu, g\u00fcvenli aral\u0131kta tutabilen bir mekanizma oldu\u011funu belirleyerek bu mekanizmaya \u201ckanalizasyon\u201d demi\u015fti. Martin\u2019e g\u00f6re e\u011fer bu kayma bir \u015fekilde bertaraf edilebilirse, bir daha b\u00fcy\u00fck resme giremeyebilecektir.<\/p>\n<p>Ara\u015ft\u0131rmalar\u0131na g\u00f6re, ya\u015flanma s\u0131ras\u0131nda gen ifadelerindeki kaymalar artmakta ve \u201cs\u00f6zde olas\u0131l\u0131ksal\u201d g\u00f6r\u00fcnen geriatrik hastal\u0131klar\u0131 olu\u015fturabilmektedir. Bu net olmakla birlikte arkas\u0131ndaki mekanizma h\u00e2l\u00e2 tam olarak bilinememektedir.<\/p>\n<p>Bu arada Washington \u00dcniver-sitesi\u2019nde ya\u015f ba\u011flant\u0131l\u0131 hastal\u0131klar\u0131 inceleyen Alexander Mendelhall, tutars\u0131zl\u0131k art\u0131\u015f\u0131na hangi h\u00fccre ya da organlar\u0131n en hassas kar\u015f\u0131l\u0131\u011f\u0131 verdi\u011fini belirleyip, \u015fansla ilk zarar g\u00f6renin ne oldu\u011funu anlamaya \u00e7al\u0131\u015f\u0131yor. Laboratuvarlar\u0131nda ba\u011f\u0131rsak kurdu olan C. elegans \u00fczerinde inceleme yap\u0131yorlar. Hangi tip h\u00fccrenin hasarland\u0131\u011f\u0131n\u0131 belirlerlerse, temel noktay\u0131 da ortaya \u00e7\u0131karabileceklerini d\u00fc\u015f\u00fcn\u00fcyorlar.<\/p>\n<p>Baz\u0131 ara\u015ft\u0131rmalar h\u00fccrelerdeki hasarlanmayla ilgilenirken, biyologlar\u0131n bir k\u0131sm\u0131 da h\u00fccre uyumu konusunda \u00e7al\u0131\u015fmalar\u0131n\u0131 y\u00fcr\u00fct\u00fcyor. Hen\u00fcz, kal\u0131tsal olmayan farkl\u0131la\u015fmalarla fayda aras\u0131ndaki sebep sonu\u00e7 ili\u015fkisi kan\u0131tlanmam\u0131\u015f olsa da, do\u011fada genetik olarak \u00f6zde\u015f topluluklarda birbirleriyle benzerlik de\u011fi\u015ftoku\u015fu oldu\u011fu g\u00f6zlenmi\u015ftir. Biyologlar buna, ekonomi d\u00fcnyas\u0131nda \u00e7ok kullan\u0131lan \u201criskten korunma\u201d diyorlar. Yani daha fazla kayb\u0131 engellemek i\u00e7in tamamen ters yat\u0131r\u0131mlara a\u00e7\u0131k olmak gibi d\u00fc\u015f\u00fcn\u00fclebilir.<\/p>\n<p>Bunun bir \u00f6rnek davran\u0131\u015f\u0131n\u0131 \u00e7ok iyi bilinen Escherichia coli bakterisinde izleyebiliyoruz. Pek \u00e7ok mikrop antibiyotiklerle \u00f6l\u00fcrken yava\u015f geli\u015fen mikroplar diren\u00e7li hale gelebiliyor. Tam olarak bilinemese de bunun bir \u201csoy devaml\u0131l\u0131\u011f\u0131\u201d sa\u011flad\u0131\u011f\u0131 d\u00fc\u015f\u00fcn\u00fclmektedir. Kanser h\u00fccrelerinde de benzer bir davran\u0131\u015f g\u00f6zlenmi\u015ftir. H\u0131zl\u0131 b\u00f6l\u00fcnen h\u00fccreler kemoterapiye daha az dayanabilmi\u015ftir.<\/p>\n<p>Mendenhall ve arkada\u015flar\u0131 da yine genetik olarak \u00f6zde\u015f C. elegans ipliksi solucan\u0131nda yapt\u0131klar\u0131 incelemede \u201c\u0131s\u0131 \u015fok proteini\u201d miktarlar\u0131ndaki farkl\u0131la\u015fmay\u0131 g\u00f6zlemlemi\u015flerdi. Bu \u00f6zel protein, di\u011fer protein yap\u0131lar\u0131 koruyarak, onlar\u0131 kanserden y\u00fcksek ate\u015fe uzanan geni\u015f bir tetikleyici aral\u0131\u011f\u0131ndaki risklerden korur. \u015eekilsiz ve kusurlu hale gelmelerini engeller.
