Biliminsanlar\u0131 konuya ili\u015fkin daha fazla \u00e7al\u0131\u015fma yap\u0131lmas\u0131 gerekti\u011finin alt\u0131n\u0131 \u00e7izerken, kan s\u0131v\u0131s\u0131 i\u00e7erisinde serbest durumda bulunan mitokondrilerin sinyal iletimi amac\u0131yla h\u00fccreler taraf\u0131ndan serbest b\u0131rak\u0131lm\u0131\u015f olabilece\u011fini d\u00fc\u015f\u00fcn\u00fcyorlar.<\/p>\n<p>Columbia \u00dcniversitesi\u2019nden Martin Picard, \u201c\u00c7ok heyecan verici, bir araya getirilen kan\u0131tlar, kan s\u0131v\u0131s\u0131 i\u00e7erisindeki partik\u00fcllerin mitokondri olduklar\u0131n\u0131 ortaya koyuyor. Yedi y\u0131l \u00f6nce Frans\u0131z Ulusal Sa\u011fl\u0131k ve T\u0131p Ara\u015ft\u0131rmalar\u0131 Enstit\u00fcs\u00fc\u2019nde kanser biyobelirte\u00e7leri \u00fczerine \u00e7al\u0131\u015fma yapan onkolog Alain Thierry \u00f6nc\u00fcl\u00fc\u011f\u00fcndeki ekip, insan kan\u0131nda dola\u015ft\u0131\u011f\u0131 bilinen serbest mitokondriyal DNA\u2019n\u0131n kanser tan\u0131s\u0131 i\u00e7in potansiyel bir i\u015faret oldu\u011funu d\u00fc\u015f\u00fcnm\u00fc\u015flerdi. Ara\u015ft\u0131rmac\u0131lar y\u00fcr\u00fctt\u00fckleri \u00e7al\u0131\u015fmalarla serbest mitokondrilere ait DNA\u2019n\u0131n, kan s\u0131v\u0131s\u0131 i\u00e7erisinde serbest bir bi\u00e7imde dola\u015fan DNA par\u00e7alar\u0131na k\u0131yasla daha uzun ve daha diren\u00e7li oldu\u011funu fark ettiler. Mitokondriyal DNA, kendi mekanizmas\u0131n\u0131 koruyan bir yap\u0131 kazanm\u0131\u015ft\u0131. Biliminsanlar\u0131 bu durumun \u00fczerine yeni deneysel \u00e7al\u0131\u015fmalara imza att\u0131lar. 80 insan\u0131n kan plazmas\u0131 incelendi. DNA diziliminin analizi i\u00e7in kullan\u0131lan tekniklerin yard\u0131m\u0131yla serbest mitokonriyal DNA\u2019n\u0131n iki ayr\u0131 tipini buldular: Bu tiplerden biri bozulmu\u015f DNA par\u00e7alar\u0131ndan olu\u015fuyordu. Di\u011feri ise olduk\u00e7a uzun zincirli ve diren\u00e7li DNA par\u00e7alar\u0131ndan olu\u015fuyordu. Biliminsanlar\u0131 bu mitokondriyal DNA\u2019lar\u0131n mitokonriyal proteinlerle ili\u015fkisini de anlamak istedi.<\/p>\n<p>Di\u011fer verileri de de\u011ferlendiren biliminsanlar\u0131, kanda serbest bir bi\u00e7imde dola\u015fan mitokondrilerin i\u015flevsel oldu\u011funu ortaya koydu. Ancak serbest mitokondrilerin i\u015flevlerinin anla\u015f\u0131lmas\u0131 i\u00e7in yeni ara\u015ft\u0131rmalar\u0131n yap\u0131lmas\u0131 gerekiyor. Serbest mitokondrilerin kan plazmas\u0131 i\u00e7indeki say\u0131s\u0131 ise 200.000 ila 3.7 milyon aras\u0131nda de\u011fi\u015febilir.<\/p>\n<p>Daha \u00f6nceki \u00e7al\u0131\u015fmalarda mitokondrilerin h\u00fccre d\u0131\u015f\u0131nda da var olabilecekleri ifade edilmi\u015fti. 2014 y\u0131l\u0131nda yap\u0131lan bir \u00e7al\u0131\u015fma kan p\u0131ht\u0131la\u015fmas\u0131 s\u0131ras\u0131nda h\u00fccrelerin mitokondrilerini kan plazmas\u0131 i\u00e7erisine b\u0131rakabileceklerini \u00f6ne s\u00fcrm\u00fc\u015ft\u00fc.
