Bu nedenle kronik hastal\u0131klar, organlarda fonksiyon kay\u0131plar\u0131 ve enfeksiyonlara kar\u015f\u0131 zay\u0131f ba\u011f\u0131\u015f\u0131kl\u0131k tepkileri ortaya \u00e7\u0131kabilir. Daha \u00f6nceki \u00e7al\u0131\u015fmalar bu s\u0131z\u0131nt\u0131lar\u0131 durdurman\u0131n ve SIRT1 enzimini yenilemenin yeni yollar\u0131 olabilece\u011fini g\u00f6stermi\u015ftir ancak yeni ara\u015ft\u0131rma, ya\u015flanma s\u00fcrecinde otofajinin rol\u00fcn\u00fc g\u00f6steren ilk \u00e7al\u0131\u015fmad\u0131r.<\/p>\n<p>SIRT1 enzimine odaklanan \u00f6nceki \u00e7al\u0131\u015fmalar, enzimin h\u00fccre metabolizmas\u0131 ve ba\u011f\u0131\u015f\u0131kl\u0131k tepkileri i\u00e7in olduk\u00e7a \u00f6nemli oldu\u011funu, enzimin -enzimin \u00fcretiminde etkili olan genin- anlat\u0131m\u0131 art\u0131r\u0131ld\u0131\u011f\u0131nda \u00f6m\u00fcr uzunlu\u011funda art\u0131\u015f oldu\u011funu g\u00f6stermekteydi. Otofaji ile SIRT1 enzimi aras\u0131ndaki ili\u015fkinin incelendi\u011fi yeni \u00e7al\u0131\u015fmada ise ara\u015ft\u0131rmac\u0131lar, ya\u015flanma s\u00fcreci (senescence) s\u0131ras\u0131nda SIRT1 enziminin aktivite kayb\u0131n\u0131n mekanizmas\u0131n\u0131 belirlemek i\u00e7in, farelerden elde edilen h\u00fccrelerde RNA dizileme tekniklerini kullanarak mRNA sentezi ve kararl\u0131l\u0131\u011f\u0131 (stabilite) ile gen anlat\u0131m\u0131n\u0131n kontrol\u00fcnde etkili olan \u00f6nemli fakt\u00f6rleri denklemden \u00e7\u0131kard\u0131. B\u00f6ylelikle otofaji ile enzim aras\u0131ndaki ili\u015fkiyi do\u011frudan inceleme f\u0131rsat\u0131 elde etti. S\u00fcreci takip eden di\u011fer deneylerde ya\u015flanm\u0131\u015f h\u00fccrelerde otofaji ile ili\u015fkili olan Atg7 proteininin mekanizmadan \u00e7\u0131kar\u0131lmas\u0131yla SIRT1 seviyelerinin yerinde kald\u0131\u011f\u0131 bulundu. B\u00f6ylece enzim kayb\u0131nda proteazomlar\u0131n de\u011fil, otofajik yolun rol oynad\u0131\u011f\u0131 g\u00f6sterildi. H\u00fccrelerdeki proteinlerin par\u00e7alanmas\u0131n\u0131 engelleyen bir proteazom inhibit\u00f6r\u00fc, ya\u015flanan h\u00fccrelerde ve dokulardaki SIRT1 proteinini geri kazanmada ba\u015far\u0131s\u0131z oldu. Aksine, otofaji inhibit\u00f6r\u00fc olan Lys05 ile ger\u00e7ekle\u015ftirilen tedavi, enzimin kayb\u0131n\u0131 \u00f6nledi. B\u00f6ylelikle enzimin lizozomlar taraf\u0131ndan par\u00e7aland\u0131\u011f\u0131 anla\u015f\u0131lm\u0131\u015f oldu.
