Fakat yeni \u00e7al\u0131\u015fmada Sibirya\u2019n\u0131n kuzeydo\u011fusundan elde edilen yeterli orandaki y\u00fcnl\u00fc mamut DNA\u2019s\u0131n\u0131n dizilenmesi ile Antik DNA \u00e7al\u0131\u015fmalar\u0131n\u0131n rekoru g\u00fcn\u00fcm\u00fczden bir milyon y\u0131l \u00f6ncesine geriledi.<\/p>\n<p>18 \u015eubat g\u00fcn\u00fc <em>Nature<\/em> dergisinde yay\u0131nlanan \u00e7al\u0131\u015fmada, 1,65 milyon, 1,34 milyon ve 0,87 milyon y\u0131ll\u0131k \u00fc\u00e7 mamut \u00f6rne\u011fi \u00fczerinden elde edilen DNA\u2019n\u0131n analizi yap\u0131ld\u0131.<\/p>\n<p>Antik DNA \u00e7al\u0131\u015fmalar\u0131nda en \u00e7ok ba\u015fvurulan \u00f6rnekler di\u015flerdir. \u00c7\u00fcnk\u00fc di\u015flerin koruyucu d\u0131\u015f tabakas\u0131, DNA\u2019n\u0131n elde edilebilece\u011fi i\u00e7yap\u0131y\u0131 olduk\u00e7a uzun bir s\u00fcre saklayabilir. Ara\u015ft\u0131rmac\u0131lar, mamutlar\u0131n di\u015flerinden DNA elde edebilmek i\u00e7in Antik DNA \u00e7al\u0131\u015fmalar\u0131nda s\u0131kl\u0131kla kullan\u0131lan birka\u00e7 y\u00f6ntemden yararland\u0131lar. Daha sonra DNA analizi ve gen dizileme \u00e7al\u0131\u015fmalar\u0131 yap\u0131p elde edilen verileri Afrika ve Asya fillerinin genomlar\u0131 ile kar\u015f\u0131la\u015ft\u0131rd\u0131lar.<\/p>\n<p>Mamut \u00f6rneklerinden ikisi, y\u00fcnl\u00fc mamuttan daha \u00f6nce gezegende var olan bir soydan geliyor gibi g\u00f6r\u00fcnmekteydi. En eski \u00f6rnek ise daha \u00f6nce tan\u0131mlanmam\u0131\u015f bir soydan geliyordu. Biliminsanlar\u0131 bu soyun, Kuzey Afrika\u2019da yay\u0131l\u0131m g\u00f6sterip, b\u00f6lgeyi kolonile\u015ftiren mamutlar\u0131n atas\u0131 oldu\u011funu d\u00fc\u015f\u00fcn\u00fcyor. Ara\u015ft\u0131rmac\u0131lar\u0131n yeni makalesi, mamutlar\u0131n evrimi hakk\u0131ndaki bilgi birikimimize katk\u0131da bulunman\u0131n yan\u0131 s\u0131ra, DNA\u2019n\u0131n d\u00fc\u015f\u00fcn\u00fclenden \u00e7ok daha eski \u00f6rneklerden bile \u00e7\u0131kar\u0131labilece\u011fini ve analiz edilebilece\u011fini g\u00f6stermektedir.<\/p>\n<p>DNA, genellikle \u00e7ok h\u0131zl\u0131 bir bi\u00e7imde bozulan, etraftaki \u00e7e\u015fitli etmenlerin etkinli\u011fi ile kontamine olabilen bir genetik materyaldir. Bu nedenle eski \u00f6rneklerden DNA elde edebilmek ve bu eski \u00f6rnekleri analiz edebilmek zordur. Daha \u00f6nceki \u00e7al\u0131\u015fmalarda, \u00f6rne\u011fin bir as\u0131rdan daha k\u0131sa bir s\u00fcre \u00f6nce nesli t\u00fckenmi\u015f olan Tilasin (Tazmanya kaplan\u0131) fosillerinin tam genom dizilimi yap\u0131lmas\u0131nda \u00e7e\u015fitli sorunlarla kar\u015f\u0131la\u015f\u0131lm\u0131\u015ft\u0131.
