Bu s\u00fcrece ili\u015fkin mekanik bilgiler, organellerin nas\u0131l ileti\u015fim kurdu\u011funa \u0131\u015f\u0131k tuttu.<\/p>\n<p>Mitokondri, h\u00fccrelerde metabolizman\u0131n ve do\u011fal ba\u011f\u0131\u015f\u0131kl\u0131k sistemi sinyallemesinin merkezi olarak hareket eden zarla \u00e7evrili organeldir. Her mitokondri, mitokondriyal genomun (mtDNA) birka\u00e7 kopyas\u0131n\u0131 i\u00e7erir ve bu mitokondriyal DNA\u2019lar d\u0131\u015far\u0131dan gelen \u00e7evresel stres fakt\u00f6rleri veya i\u00e7eriden gelen genetik mutasyonlar taraf\u0131ndan zarar g\u00f6rebilir. Bu zararlar, mtDNA\u2019n\u0131n par\u00e7alanmas\u0131na, mtDNA kopya say\u0131s\u0131n\u0131n azalmas\u0131na ve b\u00f6ylece mitokondriyal disfonksiyona (mitokondride i\u015flev bozukluklar\u0131) neden olabilir. Tigano ve di\u011ferleri h\u00fccrelerin \u00e7ekirdekte bir ba\u011f\u0131\u015f\u0131kl\u0131k tepkisi ba\u015flatmak i\u00e7in toksik mtDNA hasar\u0131n\u0131 alg\u0131lad\u0131\u011f\u0131 bir mekanizmay\u0131 ortaya \u00e7\u0131kar\u0131r.<\/p>\n<p>Viral enfeksiyon veya \u0131\u015f\u0131nlaman (irradiyasyon) gibi akut stres ko\u015fullar\u0131, h\u00fccrede apoptotik (programl\u0131 h\u00fccre \u00f6l\u00fcm\u00fc) yollar\u0131n aktivasyonuna yol a\u00e7abilir. Mitokondri bu apoptotik yollarda \u00f6nemli bir role sahiptir. BAK ve BAX ad\u0131 verilen g\u00f6zenek olu\u015fturan proteinler mitokondriyal zarda birikerek, h\u00fccre \u00f6l\u00fcm fakt\u00f6rlerinin organelden h\u00fccrenin sitozolik s\u0131v\u0131s\u0131na sal\u0131nmas\u0131na yol a\u00e7ar ve bu s\u00fcrece herniyasyon ad\u0131 verilir. Baz\u0131 durumlarda, h\u00fccre \u00f6l\u00fcm fakt\u00f6rleri aktif de\u011fildir, bu durumda h\u00fccre \u00f6l\u00fcm fakt\u00f6rleri yerine DNA ve RNA gibi mitokondriyal i\u00e7erikler sitozole sal\u0131n\u0131r. Sitozolde mtDNA ve mtRNA birikmesi g\u00fc\u00e7l\u00fc bir antiviral tepki ba\u015flat\u0131r. Ancak, mitokondriyal disfonksiyonun tam olarak hangi y\u00f6nlerinin bu mitokondriyal materyalin ekstr\u00fczyonuna (sitozola sal\u0131nmas\u0131na) ve birikmesine yol a\u00e7t\u0131\u011f\u0131 a\u00e7\u0131k de\u011fildir.<\/p>\n<p>Tigano ve di\u011ferleri h\u00fccre i\u00e7in bir stres bi\u00e7imi olan mtDNA\u2019n\u0131n kesilmesini incelemek i\u00e7in yola \u00e7\u0131kt\u0131lar. Grup, mtDNA\u2019n\u0131n her iki sarmal\u0131nda da kesikler olu\u015fturan mitokondri hedefli TALEN\u2019ler (mtTLN) ad\u0131 verilen &#8220;molek\u00fcler makas&#8221; yap\u0131lar\u0131n\u0131 kullanarak memeli h\u00fccrelerini manip\u00fcle ettiler. TALEN molek\u00fcl\u00fc DNA\u2019n\u0131n istenilen sekans\u0131ndan \u00e7ift sarmall\u0131 kesikler olu\u015fturabilen bir enzimdir. mtTLN ise mitokondriyal DNA\u2019y\u0131 kesmesi i\u00e7in modifiye edilmi\u015f bir TALEN enzimidir. mtTLN uygulanan h\u00fccreler ve mtTLN uygulanmayan h\u00fccreler aras\u0131ndaki gen ifade de\u011fi\u015fikliklerini analiz edebilmek i\u00e7in RNA dizileme tekni\u011fini kulland\u0131lar. Analizin sonucunda, interferon gibi genellikle viral enfeksiyonlarla sava\u015f\u0131rken kar\u015f\u0131la\u015ft\u0131\u011f\u0131m\u0131z do\u011fal ba\u011f\u0131\u015f\u0131kl\u0131k tepkisinde yer alan \u00e7ekirdek genlerinin ifadesinin artt\u0131\u011f\u0131n\u0131 buldular. Yazarlar ayr\u0131ca STAT1 ad\u0131 verilen transkripsiyon fakt\u00f6r\u00fcn\u00fcn fosfat gruplar\u0131 taraf\u0131ndan modifiye edildi\u011fini ve interferon tepkisinde kritik bir \u00f6nemi olan STAT1\u2019in \u00e7ekirde\u011fe ta\u015f\u0131nmas\u0131 olay\u0131n\u0131n ger\u00e7ekle\u015fti\u011fini buldular.<\/p>\n<p>Toksik, DNA\u2019ya zarar veren ajanlara maruz b\u0131rak\u0131lma yoluyla ya da DNA\u2019n\u0131n kendi kopyas\u0131n\u0131 olu\u015ftururken ger\u00e7ekle\u015fen hatalarla meydana gelen mtDNA\u2019daki k\u0131r\u0131lmalar genellikle organel i\u015flevinin bozulmas\u0131na neden olur. Ancak Tigano ve meslekta\u015flar\u0131 mtTLN uygulanan h\u00fccrelerde mtDNA say\u0131s\u0131n\u0131n y\u00fczde 60 oran\u0131nda azald\u0131\u011f\u0131n\u0131 ve bunun mitokondriyal fonksiyon \u00fczerinde hemen bir etkisi olmad\u0131\u011f\u0131n\u0131 buldu. Grup, morfoloji, mitokondri zar\u0131nda olan proton (H+ iyonlar\u0131) de\u011fi\u015fiminde ve reaktif oksijen t\u00fcrlerinin olu\u015fturmas\u0131 gibi normal mitokondriyal fonksiyonun temel g\u00f6stergelerinde de\u011fi\u015fiklik g\u00f6zlemlemedi. Bu veriler, mtDNA kesilmesinin antiviral tepkilerin anahtar tetikleyicisi oldu\u011funu g\u00f6stermektedir.<\/p>\n<p>Sonra, Tigano ve arkada\u015flar\u0131 mtDNA kesilmesini \u00e7ekirde\u011fe mesaj olarak ileten sinyal molek\u00fcllerini tan\u0131mlamaya giri\u015ftiler. mtTLN uygulanan h\u00fccrelerin sa\u011flam mitokondriyal fonksiyonu olmas\u0131na ve apoptotik olmamas\u0131na ra\u011fmen, mitokondriyal herniyasyonla uyumlu olarak mitokondriyal zar\u0131n \u00fczerinde BAK-BAX taraf\u0131ndan olu\u015fturulan g\u00f6zeneklerin olu\u015ftu\u011funu g\u00f6sterdiler. Ara\u015ft\u0131rmac\u0131lar, mtDNA\u2019n\u0131n mitokondride kal\u0131rken mtRNA\u2019n\u0131n bu h\u00fccrelerin sitozol\u00fcnde birikmi\u015f oldu\u011funu buldular. mtRNA molek\u00fclleri, RIG-I ad\u0131 verilen sitozoldeki viral RNA sens\u00f6r\u00fc olarak bilinen bir RNA alg\u0131lay\u0131c\u0131 protein taraf\u0131ndan tespit edildi. Mitokondriyal antiviral sinyalleme (MAVS) olarak adland\u0131r\u0131lan mitokondriyal d\u0131\u015f zar \u00fczerindeki adapt\u00f6r proteini ile \u00e7al\u0131\u015fan RIG-I, \u00e7ekirdekteki interferon yan\u0131t genlerini etkinle\u015ftiren bir sinyal yolunu tetikler. Bu uygulama mtRNA\u2019n\u0131n, BAK-BAX proteinlerinin olu\u015fturdu\u011fu g\u00f6zeneklerden ge\u00e7erek sitozola sal\u0131nmas\u0131n\u0131 tetikler. Sitozolik RNA sens\u00f6r\u00fc RIG-I ve birlikte \u00e7al\u0131\u015ft\u0131\u011f\u0131 adapt\u00f6r\u00fc MAVS sitozoldaki mtRNA\u2019y\u0131 alg\u0131lar ve interferon tepkisinde yer alan ba\u011f\u0131\u015f\u0131kl\u0131k sistemi genlerinin ifadesinin artmas\u0131na sebep olur. Buna ek olarak, STAT1 transkripsiyon fakt\u00f6r\u00fc fosfatlan\u0131r ve interferon tepki genlerini aktifle\u015ftirmede yer alma ihtimali olan STAT1 \u00e7ekirdek i\u00e7ine ta\u015f\u0131n\u0131r.<\/figcaption><\/figure>\n<p>Kanser tedavisinde kullan\u0131lan radyasyon gibi DNA hasar\u0131 olu\u015fturan fakt\u00f6rler \u00e7ekirdekteki DNA hasar\u0131ndan kaynakland\u0131\u011f\u0131 d\u00fc\u015f\u00fcn\u00fclen sistemik bir ba\u011f\u0131\u015f\u0131kl\u0131k tepkisi ortaya \u00e7\u0131kar\u0131r Tigano ve meslekta\u015flar\u0131, radyasyonun mtDNA say\u0131lar\u0131n\u0131 y\u00fczde 40 oran\u0131nda azaltt\u0131\u011f\u0131n\u0131 ve mtTLN&#8217;lerle ayn\u0131 ba\u011f\u0131\u015f\u0131kl\u0131k tepkisini ortaya \u00e7\u0131kard\u0131\u011f\u0131n\u0131 buldular, bu da \u00e7ift sarmall\u0131 kesiklerin mtDNA&#8217;da oldu\u011fu kadar \u0131\u015f\u0131nlamadan sonra h\u00fccre \u00e7ekirde\u011findeki DNA&#8217;da da meydana geldi\u011fini \u00f6ne s\u00fcr\u00fcyor. \u00c7arp\u0131c\u0131 bir \u015fekilde, \u0131\u015f\u0131nlama s\u0131ras\u0131nda interferon tepkisinin uyar\u0131lmas\u0131, mtDNA&#8217;s\u0131 olmayan h\u00fccrelerde neredeyse tamamen iptal edildi. Bu g\u00f6zlem, radyasyonun neden oldu\u011fu mtDNA hasar\u0131n\u0131n interferon tepkilerinin tetikleyicisi olabilece\u011fini g\u00f6stermektedir. Dikkat \u00e7ekici bir \u015fekilde, di\u011fer birka\u00e7 do\u011fal ba\u011f\u0131\u015f\u0131kl\u0131k tepkilerinin uyar\u0131lmas\u0131, mtDNA&#8217;s\u0131 olmayan h\u00fccrelerde devam etti, bu da mtDNA&#8217;n\u0131n olmay\u0131\u015f\u0131n\u0131n \u00f6zellikle interferon tepkisini bozdu\u011funu d\u00fc\u015f\u00fcnd\u00fcr\u00fcyor.<\/p>\n<p>\u00c7al\u0131\u015fma, mtRNA\u2019n\u0131n ba\u011f\u0131\u015f\u0131kl\u0131k sistemi uyar\u0131c\u0131 bir rol\u00fc olabilece\u011fini vurgulamaktad\u0131r. Ancak yine de cevaplanmas\u0131 gereken sorular bulunuyor. \u00d6rne\u011fin, mtRNA molek\u00fclleri do\u011fas\u0131 gere\u011fi olduk\u00e7a karars\u0131z bir yap\u0131ya sahiptirler, peki nas\u0131l oluyor da kararl\u0131 bir hale getirilip sitozolde par\u00e7alanmadan birikebiliyorlar? Gelecekteki ara\u015ft\u0131rmalar i\u00e7in bir ba\u015fka konu ise mtDNA kesildikten sonra BAK-BAX g\u00f6zeneklerinin olu\u015fumunu tetikleyen fakt\u00f6rlerin neler oldu\u011fudur. Bu g\u00f6zenek olu\u015fumunu engelleyen ila\u00e7lar\u0131n interferon ba\u011f\u0131\u015f\u0131kl\u0131k tepkisini bask\u0131lay\u0131p bask\u0131layamayaca\u011f\u0131n\u0131 ara\u015ft\u0131rmak geni\u015f bir ilgi yaratacakt\u0131r. H\u00fccrelerin bir ba\u011f\u0131\u015f\u0131kl\u0131k tepkisi ba\u015flatmak i\u00e7in mitokondriden kendi RNA&#8217;lar\u0131n\u0131 tan\u0131d\u0131\u011f\u0131 bir mekanizman\u0131n ke\u015ffi, bu yolun otoimm\u00fcn hastal\u0131\u011fa dahil olup olmad\u0131\u011f\u0131 sorusunu da g\u00fcndeme getiriyor.
