Fakat sa\u011fl\u0131kl\u0131 insanlarda ba\u011f\u0131\u015f\u0131kl\u0131k tolerans\u0131 olarak bilinen bir s\u00fcre\u00e7 i\u015f g\u00f6r\u00fcr ve otoimm\u00fcn hastal\u0131klara do\u011fru giden s\u00fcre\u00e7 engellenebilir. Ba\u011f\u0131\u015f\u0131kl\u0131k tolerans\u0131n\u0131n v\u00fccudun patojenlerle etkili bir bi\u00e7imde m\u00fccadele etmesini engellememesi i\u00e7in \u00e7ok say\u0131da karma\u015f\u0131k mekanizma, ba\u011f\u0131\u015f\u0131kl\u0131k yan\u0131t\u0131 ile bask\u0131lama mekanizmas\u0131 aras\u0131ndaki dengeyi sa\u011flamak i\u00e7in \u00e7al\u0131\u015f\u0131r.<\/p>\n<p><em>PNAS<\/em>\u2019ta yay\u0131nlanan bir \u00e7al\u0131\u015fma, bu dengeyi korumaya yard\u0131mc\u0131 olan iki resept\u00f6r\u00fc ve bu iki resept\u00f6r aras\u0131ndaki ili\u015fkinin s\u00f6z konusu dengeye etkisini ortaya \u00e7\u0131kard\u0131. DNAM-1 ve TIGIT ad\u0131 verilen bu iki resept\u00f6r daha \u00f6nce de baz\u0131 ba\u011f\u0131\u015f\u0131kl\u0131k h\u00fccrelerindeki (T h\u00fccreleri veya Treg h\u00fccreleri) fonksiyonlar\u0131 bak\u0131m\u0131ndan analiz edilmi\u015fti.<\/p>\n<p>Treg h\u00fccreleri, ba\u011f\u0131\u015f\u0131kl\u0131k tolerans\u0131 ve otoimm\u00fcn yan\u0131t\u0131n \u00f6nlenmesi konusunda \u00f6nemli olan arac\u0131 yap\u0131lard\u0131r. V\u00fccudun istilac\u0131 ile etkili bir bi\u00e7imde sava\u015fmas\u0131n\u0131 ve onu temizlemesini sa\u011flayan ba\u011f\u0131\u015f\u0131kl\u0131k reaksiyonlar\u0131 s\u0131ras\u0131nda Treg h\u00fccrelerinin aktivitesi bask\u0131lan\u0131r. Ancak Treg h\u00fccrelerindeki bu dengeyi kontrol eden molek\u00fcler mekanizmalar tam olarak anla\u015f\u0131lamam\u0131\u015ft\u0131.<\/p>\n<p>Yeni \u00e7al\u0131\u015fmada dikkat \u00e7ekildi\u011fi \u00fczere aktifle\u015ftirici etkisi olan CD155 molek\u00fcl\u00fc DNAM-1 ve TIGIT resept\u00f6rlerine ba\u011fland\u0131\u011f\u0131nda, T h\u00fccrelerine nas\u0131l davranacaklar\u0131n\u0131 ve hangi i\u015flevleri yerine getireceklerini s\u00f6yleyen sinyalleri tetikler. Ekip, DNAM-1 ve TGIT resept\u00f6rlerinin enfeksiyon varl\u0131\u011f\u0131nda CD155 i\u00e7in rekabete girdi\u011fini raporlad\u0131. Buna g\u00f6re CD155, DNAM-1&#8217;e ba\u011fland\u0131\u011f\u0131nda, ba\u011f\u0131\u015f\u0131kl\u0131k sistemine onu harekete ge\u00e7irici mesajlar g\u00f6nderilir. Fakat CD155, TIGIT&#8217;e ba\u011fland\u0131\u011f\u0131nda, s\u00fcre\u00e7 ters y\u00f6nde i\u015fler ve ba\u011f\u0131\u015f\u0131kl\u0131k sistemi bask\u0131lan\u0131r; b\u00f6ylece T h\u00fccreleri ba\u011f\u0131\u015f\u0131kl\u0131k yan\u0131t\u0131 olu\u015fturmada i\u015f g\u00f6remez.<\/p>\n<p>Ara\u015ft\u0131rmac\u0131lar DNAM-1 ve TIGIT aras\u0131ndaki molek\u00fcler etkile\u015fimleri ara\u015ft\u0131rmak i\u00e7in \u00e7e\u015fitli deneyler yapt\u0131. DNAM-1 resept\u00f6r\u00fc ta\u015f\u0131mayan deneklerde geli\u015fmi\u015f d\u00fczeyde TIGIT arac\u0131l\u0131 sinyalleme oldu\u011fu g\u00f6zlendi. Bu durumun temel nedeni DNAM-1 mevcut olmad\u0131\u011f\u0131nda TIGIT\u2019e ba\u011flanmak i\u00e7in ortamda daha fazla CD155\u2019in kalmas\u0131d\u0131r. B\u00f6ylece ba\u011f\u0131\u015f\u0131kl\u0131\u011f\u0131n bask\u0131lanmas\u0131 uzun bir s\u00fcre devam eder.<\/p>\n<p>Treg&#8217;lerin otoimm\u00fcn hastal\u0131klar i\u00e7in \u00f6nemli bir rol oynad\u0131\u011f\u0131 d\u00fc\u015f\u00fcn\u00fcld\u00fc\u011f\u00fcnde, ara\u015ft\u0131rman\u0131n sonu\u00e7lar\u0131, otoimm\u00fcn hastal\u0131\u011f\u0131 olan bireylerde DNAM-1 ve TIGIT aras\u0131ndaki bozulmu\u015f dengenin yeniden kurulabilece\u011fini g\u00f6stermektedir.
