Yeni \u00e7al\u0131\u015fmaya g\u00f6re bu ba\u011f dokusu proteini, sepsis denilen ve \u00f6l\u00fcmc\u00fcl olabilen tepkinin k\u0131s\u0131tlanmas\u0131na da yard\u0131mc\u0131 oluyor.<\/p>\n<p>Ba\u011f dokusu h\u00fccrelerinin h\u00fccreler aras\u0131 etkile\u015fimlerde sinyal ortakla\u015fmas\u0131n\u0131 sa\u011flayan h\u00fccre d\u0131\u015f\u0131 matrise (ECM) odaklanan \u00e7al\u0131\u015fmalar, sinyallerin herhangi bir sebeple i\u015f g\u00f6rmemesinin hastal\u0131klara neden oldu\u011funu g\u00f6stermi\u015fti. ECM&#8217;deki \u00f6nemli yap\u0131lar aras\u0131nda kolajen gibi sert yap\u0131sal matris proteinleri \u00fcreten fibroblastlar bulunmaktad\u0131r.<\/p>\n<p>Yeni \u00e7al\u0131\u015fma ise fibroblastlar taraf\u0131ndan salg\u0131lanan ve ba\u011f dokular\u0131nda kolajen ile ortakl\u0131k kurdu\u011fu bilinen bir protein-\u015feker kombinasyonu (proteoglikan) olan lumikan\u0131n ba\u011f\u0131\u015f\u0131kl\u0131k sistemi ile ili\u015fkisine odaklanmaktad\u0131r. \u00c7al\u0131\u015fmaya g\u00f6re lumikan, makrofaj h\u00fccreleri \u00fczerinden ba\u011f\u0131\u015f\u0131kl\u0131k sisteminin \u00fcretti\u011fi tepkiyi desteklemektedir. <em>Proceedings of the National Academy of Sciences<\/em> (<em>PNAS<\/em>) portal\u0131nda yay\u0131nlanan \u00e7al\u0131\u015fmaya g\u00f6re lumikan, bir vir\u00fcse veya \u00f6l\u00fcrken DNA\u2019lar\u0131n\u0131 ortama b\u0131rakan bir h\u00fccreye (dokular\u0131n stres alt\u0131nda oldu\u011funu g\u00f6sterir) kar\u015f\u0131 ba\u011f\u0131\u015f\u0131kl\u0131k yan\u0131t\u0131n\u0131n s\u0131n\u0131rlanmas\u0131na yard\u0131mc\u0131 olarak dokular\u0131 korur. Bu t\u00fcr ba\u011f\u0131\u015f\u0131kl\u0131k yan\u0131tlar\u0131 iyile\u015fme a\u015famas\u0131na ge\u00e7i\u015fi temsil eder, fakat tepkiler sepsiste \u00e7ok b\u00fcy\u00fcr ve v\u00fccudun kendi dokular\u0131na zarar vermesine neden olur.<\/p>\n<p>Biliminsanlar\u0131, lumikan\u0131n, enfeksiyon ajanlar\u0131yla ili\u015fkili molek\u00fcllere ba\u011flanabilen resept\u00f6rlerin aktivitesini kontrol eden ba\u011f\u0131\u015f\u0131kl\u0131k h\u00fccrelerinin y\u00fczey proteinleriyle etkile\u015fime girebildi\u011fini g\u00f6sterdi. Bu resept\u00f6rler, a\u015f\u0131r\u0131 aktifle\u015fmeleriyle birlikte ba\u011f\u0131\u015f\u0131kl\u0131k h\u00fccrelerinin v\u00fccudun kendi dokular\u0131na sald\u0131rmas\u0131na da neden olabilir.<\/p>\n<p>Lumikan\u0131n ba\u011f\u0131\u015f\u0131kl\u0131k yan\u0131t\u0131 \u00fcreten h\u00fccrelerin y\u00fczeylerindeki etkisinin tan\u0131mlanmas\u0131n\u0131n yan\u0131 s\u0131ra, bu \u00e7al\u0131\u015fmada, lumikan\u0131n endozom ad\u0131 verilen h\u00fccre zar\u0131na ba\u011fl\u0131 keselerle h\u00fccre i\u00e7ine do\u011fru \u00e7ekildi\u011fi de g\u00f6sterildi. Lumikan\u0131n h\u00fccre d\u0131\u015f\u0131ndaki etkisi ile h\u00fccre d\u0131\u015f\u0131ndaki etkisi birbirinin tersine gibi g\u00f6r\u00fcnse de her iki aktivitenin de ba\u011f\u0131\u015f\u0131kl\u0131k sisteminin tepkisini g\u00fc\u00e7lendirici rol\u00fc bulunmaktad\u0131r.<\/p>\n<p>Deneyler ve analizler, lumikan \u00fcreten genden yoksun olacak \u015fekilde tasarlanan farelerde hem bakteriyel enfeksiyonlarla m\u00fccadelede hem de sepsise kar\u015f\u0131 diren\u00e7te \u00e7e\u015fitli sorunlar ortaya \u00e7\u0131kt\u0131\u011f\u0131n\u0131 g\u00f6sterdi.
