Kopyalanan ilk canl\u0131 Dolly adl\u0131 bir koyundu. Dolly\u2019nin ard\u0131ndan onlarca \u00fclkede, y\u00fczlerce hayvan kopyaland\u0131. Kopyalama konusunda T\u00fcrkiye\u2019de de T\u00dcB\u0130TAK Gen M\u00fchendisli\u011fi ve Biyoteknoloji Ara\u015ft\u0131rma Enstit\u00fcs\u00fc\u2019nde (GMBAE) baz\u0131 \u00e7al\u0131\u015fmalar yap\u0131l\u0131yor.\u00a0 Bu \u00e7al\u0131\u015fmalar\u0131 T\u00dcB\u0130TAK-GMBAE ara\u015ft\u0131rmac\u0131s\u0131 Do\u00e7. Dr. Sezen Arat \u015f\u00f6yle anlat\u0131yor:<\/p>\n<p>Kopyalama, \u00f6teki ad\u0131yla klonlama, bir canl\u0131n\u0131n t\u0131pat\u0131p ayn\u0131s\u0131 (genetik yap\u0131s\u0131 ayn\u0131 olan) canl\u0131lar\u0131n olu\u015fturulmas\u0131 anlam\u0131na gelir ve \u00fcremenin e\u015feysiz bi\u00e7midir. Memelilerde kopyalama \u00e7al\u0131\u015fmalar\u0131, 1980\u2019li y\u0131llar\u0131n ba\u015f\u0131ndan beri y\u00fcr\u00fct\u00fcl\u00fcyor. Memelilerde d\u00f6llenmi\u015f yumurtan\u0131n anne rahminde b\u00f6l\u00fcnmesi ve ayr\u0131 ayr\u0131 geli\u015fmesi sonucu olu\u015fan tek yumurta ikizleri de asl\u0131nda do\u011fal kopyalard\u0131r. Bu nedenle ilk klonlama \u00e7al\u0131\u015fmalar\u0131, bu do\u011fal olay\u0131n kopya edilmesiyle ba\u015flad\u0131. Burada uygulanan y\u00f6ntem, bir embriyonun h\u00fccrelerini da\u011f\u0131tarak ayr\u0131 ayr\u0131 geli\u015fmesini sa\u011flamakt\u0131. B\u00f6ylece bir embriyondan birden fazla embriyon \u00fcretilebiliyordu.<\/p>\n<p>Bir canl\u0131n\u0131n ger\u00e7ek anlamda ilk kopyas\u0131 1997\u2019de yap\u0131labilmi\u015ftir. Bilindi\u011fi gibi, ilk kopya memeli 6 ya\u015f\u0131ndaki bir koyunun kopyas\u0131 olan Dolly\u2019dir. Bu \u00e7al\u0131\u015fma, bir d\u00f6n\u00fcm noktas\u0131 olmu\u015ftur. Bunu izleyen 7 y\u0131l i\u00e7inde, s\u0131\u011f\u0131r, fare, ke\u00e7i, domuz, tav\u015fan, kedi, kat\u0131r ve at bu y\u00f6ntemle kopyaland\u0131. Bu teknolojinin \u00e7e\u015fitli uygulama alanlar\u0131 ve \u00e7ok geni\u015f bir a\u00e7\u0131l\u0131m\u0131 vard\u0131r. Bir canl\u0131y\u0131 \u00e7o\u011faltabilirsiniz; \u00f6lm\u00fc\u015f bir canl\u0131y\u0131 yeniden do\u011faya kazand\u0131rabilirsiniz ya da de\u011fi\u015fik ama\u00e7larla (\u00f6rne\u011fin, verim \u00f6zelliklerini iyile\u015ftirmek, hastal\u0131klara diren\u00e7 kazand\u0131rmak, organ kayna\u011f\u0131 yaratmak vb. i\u00e7in) gen yap\u0131s\u0131 de\u011fi\u015ftirilmi\u015f hayvanlar olu\u015fturabilirsiniz.<\/p>\n<p>T\u00fcrkiye, bu alanda ad\u0131n\u0131, 2001 y\u0131l\u0131ndan bu yana bilim d\u00fcnyas\u0131nda duyurmaya ba\u015flad\u0131. Kopyalamayla ilgili uluslararas\u0131 dergilerde, T\u00dcB\u0130TAK-GMBAE adresini ta\u015f\u0131yan 15\u2019in \u00fczerinde yay\u0131n var. \u00dclkemizde kopyalamaya y\u00f6nelik ilk \u00e7al\u0131\u015fmalar Eyl\u00fcl 2002\u2019de T\u00dcB\u0130TAK-GMBAE\u2019de ba\u015flad\u0131. Bu projede ilk ama\u00e7, d\u00fcnyadaki teknolojiyi \u00fclkemize transfer etmekti. \u0130kinci ama\u00e7sa, bir canl\u0131 \u00f6ld\u00fckten sonra h\u00fccrelerinin so\u011fukta canl\u0131l\u0131\u011f\u0131n\u0131 ve kopyalamada kullan\u0131labilirli\u011fini ne kadar s\u00fcre koruyabilece\u011fini belirlemekti. \u00c7al\u0131\u015fmam\u0131zda, 2 ve 3 g\u00fcn \u00f6nce \u00f6lm\u00fc\u015f ve so\u011fuk depoda korunan s\u0131\u011f\u0131r dokular\u0131ndan h\u00fccre elde ettik. Sonra da bu h\u00fccreleri kullanarak laboratuvar\u0131m\u0131zda kopya s\u0131\u011f\u0131r embriyonlar\u0131 geli\u015ftirdik. Bu \u00e7al\u0131\u015fma, Ocak 2004\u2019te ABD\u2019de yap\u0131lan Uluslararas\u0131 Embriyon Transferi Kongresi\u2019nde sunuldu. \u0130kinci \u00e7al\u0131\u015fma, \u00f6n\u00fcm\u00fczdeki y\u0131l Danimarka\u2019da yap\u0131lacak bir ba\u015fka uluslararas\u0131 kongrede sunulacak.<\/p>\n<p>Bu projenin ard\u0131ndan, Kas\u0131m 2003\u2019te Macar Bilimler Akademisi ile ortak bir kopyalama \u00e7al\u0131\u015fmas\u0131na ba\u015flad\u0131k. Bu projenin ilk amac\u0131, kopya s\u0131\u011f\u0131r embriyonlar\u0131n\u0131 dondurarak saklamak. \u0130kinci ama\u00e7sa bu embriyonlar\u0131 transfer ederek, T\u00fcrkiye\u2019de ilk kez kopya bir s\u0131\u011f\u0131r \u00fcretmek. Bunun i\u00e7in yerli \u0131rklar\u0131m\u0131zdan birini kopyalamay\u0131 ama\u00e7l\u0131yoruz. Embriyon transferlerine bu y\u0131l\u0131n sonunda ba\u015flamay\u0131 planl\u0131yoruz. E\u011fer her \u015fey yolunda giderse 2005 y\u0131l\u0131n\u0131n sonlar\u0131nda kopya s\u0131\u011f\u0131rlar\u0131n do\u011faca\u011f\u0131n\u0131 umut ediyoruz. Enstit\u00fcn\u00fcn arazisi i\u00e7inde bir b\u00fcy\u00fckba\u015f hayvan deney \u00e7iftli\u011fi olmad\u0131\u011f\u0131 i\u00e7in, embriyon transferleri Uluda\u011f \u00dcniversitesi\u2019nin deney hayvan\u0131 \u00e7iftli\u011finde ger\u00e7ekle\u015ftirilecek. Bu projelerde s\u0131\u011f\u0131r\u0131n se\u00e7ilmesinin nedeni; s\u0131\u011f\u0131r\u0131n, halk\u0131m\u0131z\u0131n k\u0131rm\u0131z\u0131 et gereksiniminin b\u00fcy\u00fck bir b\u00f6l\u00fcm\u00fcn\u00fc kar\u015f\u0131layan ve ayn\u0131 zamanda ekonomik de\u011feri y\u00fcksek bir t\u00fcr olmas\u0131d\u0131r. D\u00fcnyada da kopyalama alan\u0131nda \u00fczerinde en \u00e7ok \u00e7al\u0131\u015f\u0131lan t\u00fcr s\u0131\u011f\u0131rd\u0131r.<\/p>\n<p>Kopyalama teknolojisinde bir\u00e7ok t\u00fcrde ba\u015far\u0131ya ula\u015f\u0131lmas\u0131na kar\u015f\u0131n, h\u00e2l\u00e2 \u00e7\u00f6z\u00fclmesi gereken baz\u0131 sorunlar vard\u0131r. Daha geli\u015fimini tamamlamam\u0131\u015f bir teknolojidir. Kopyalamada kar\u015f\u0131la\u015f\u0131lan ba\u015fl\u0131ca sorunlar; transfer edilen 100 kopya embriyondan yaln\u0131zca 15-20 tanesinin sa\u011fl\u0131kl\u0131 bir canl\u0131 olu\u015fturabilmesi, baz\u0131 kopyalarda dola\u015f\u0131m bozukluklar\u0131, solunum yetersizlikleri ve ba\u011f\u0131\u015f\u0131kl\u0131k sistemi bozukluklar\u0131 g\u00f6r\u00fclmesidir. Bunlar\u0131n nedenleri hen\u00fcz anla\u015f\u0131lamam\u0131\u015ft\u0131r. \u00d6te yandan, bug\u00fcne de\u011fin y\u00fczlerce sa\u011fl\u0131kl\u0131 kopya hayvan da \u00fcretilmi\u015ftir. \u00dczerinde \u00e7al\u0131\u015f\u0131lan ve endi\u015fe uyand\u0131ran bir ba\u015fka konu da do\u011fan kopyalar\u0131n, kopyalanan canl\u0131n\u0131n ya\u015f\u0131nda do\u011fma olas\u0131l\u0131\u011f\u0131n\u0131n bulunmas\u0131 ve bu y\u00fczden de daha k\u0131sa ya\u015fayabilecek olmas\u0131d\u0131r. Ancak yap\u0131lan bir\u00e7ok \u00e7al\u0131\u015fma, bunun her ko\u015fulda olmad\u0131\u011f\u0131n\u0131, bu nedenle de ger\u00e7ekte \u00f6nemli bir sorun olu\u015fturmad\u0131\u011f\u0131n\u0131 g\u00f6stermektedir. Yarat\u0131c\u0131lar\u0131, Dolly\u2019nin \u00f6l\u00fcm\u00fcn\u00fcn, teknolojideki bir hatadan kaynaklanmad\u0131\u011f\u0131n\u0131, bu \u00f6l\u00fcm\u00fcn ayn\u0131 d\u00f6nemde ba\u015fka bir\u00e7ok normal koyunun da \u00f6l\u00fcm\u00fcne neden olan bir enfeksiyon hastal\u0131k y\u00fcz\u00fcnden oldu\u011funu a\u00e7\u0131klam\u0131\u015flard\u0131r.<\/p>\n<p>Elbetteki kopyalamayla ilgili birtak\u0131m sorunlar\u0131n bizim kopyalar\u0131m\u0131zda da ortaya \u00e7\u0131kma olas\u0131l\u0131\u011f\u0131 vard\u0131r. Bunlar embriyon \u00e7al\u0131\u015fmalar\u0131n\u0131n hepsinde kar\u015f\u0131la\u015f\u0131labilecek sorunlard\u0131r. Ekip olarak bunlarla kar\u015f\u0131la\u015fma riskini en aza indirecek \u00f6nlemleri almaya \u00e7al\u0131\u015f\u0131yoruz.
